Goldenrod Therapeutics Secures $6.5 Million Seed Round to Advance CNS-Penetrant PDE4 Inhibitor Toward 2027 Clinical Trials

A startup built on research from the University of Nebraska Medical Center has raised $6.5 million in seed financing to propel a novel brain-penetrant PDE4 (搜索) inhibitor toward the clinic. Goldenrod Therapeutics (搜索), established in 2023 with support from UNeMed (搜索), UNMC's technology transfer office, announced the funding round will support the studies required to initiate clinical trials in 2027.
The company's lead asset, designated 11h (搜索), was developed by Corey Hopkins, PhD, at the UNMC College of Pharmacy. The molecule targets phosphodiesterase 4 (PDE4 (搜索)), an enzyme linked to a broad spectrum of inflammatory and central nervous system disorders.
A Decades-Old Challenge in PDE4 (搜索) Inhibition
PDE4 (搜索) has long been recognized as a therapeutic target in inflammatory diseases including chronic obstructive pulmonary disease (搜索), rheumatoid arthritis (搜索), and inflammatory bowel disease (搜索), as well as CNS conditions such as schizophrenia (搜索), drug addiction, Alzheimer's disease (搜索), multiple sclerosis (搜索), ALS (搜索), and Batten disease (搜索). However, existing PDE4-targeting medications have been hampered by two persistent limitations: significant side effects—particularly nausea and vomiting—and poor penetration into the central nervous system.
Dr. Hopkins' compound appears to surmount both obstacles. According to UNMC, the molecule demonstrated reduced potential for adverse side effects while showing dramatic improvement in brain uptake compared to currently available PDE4 (搜索) inhibitors.
"This is what world-class drug discovery looks like, and it is happening right here at UNMC," said Michael Dixon, PhD, President and CEO of UNeMed (搜索). "Dr. Hopkins and his team have designed a molecule that overcomes a problem the pharmaceutical industry wrestled with for decades, and now a company has the capital to carry it into the clinic."
From Academic Lab to Startup
Goldenrod Therapeutics (搜索) was formed in 2023 with UNeMed (搜索) structuring the licensing framework for the technology's use and development. The seed round was led by Ataxia Ventures (搜索) and an affiliate of Fannin Partners (搜索).
"The scientific foundation established at UNMC and Fannin has positioned 11h (搜索) as a potentially important advancement in CNS-targeted PDE4 (搜索) inhibition, with potential relevance across a broader range of diseases," said Tom Hamilton of Ataxia Ventures (搜索).
Dr. Hopkins, reflecting on the molecule's trajectory, emphasized the collective effort behind its development. "It is exciting and gratifying to see your work possibly improve someone's life. For 11h (搜索), its progress means a lot because of all the effort put in by students and postdocs over the years. So, it's fantastic that their hard work is getting closer to actually helping patients."
He added: "One of the things I love most about drug discovery is that you never know where a promising molecule might end up helping the most. This condition desperately needs new treatments, so it's amazing to be closing that gap."
Institutional Investment in Drug Discovery
The financing underscores UNMC's growing role in translational research. "Drug discovery is some of the most important work we do at UNMC, and investors place real value on new molecules and discoveries," Dixon said. "A raise like this shows what our science is worth, and the Center for Drug Design and Innovation is at the heart of that effort. As our research enterprise grows, continued investment in drug discovery and development will be critical to our success creating and developing new therapeutics."
With the seed funding secured, Goldenrod Therapeutics (搜索) will now focus on completing the preclinical and IND-enabling studies necessary to enter human trials, with a target clinical start date of 2027.
