Indena and TCG GreenChem Form Strategic Alliance to Accelerate ADC Payload-Linker Development
核心洞察
Indena and TCG GreenChem (搜索) have announced a strategic collaboration to develop innovative payload-linkers (搜索) for antibody-drug conjugates (搜索), combining Indena's high-potency API expertise with TCG GreenChem's green chemistry capabilities.
The partnership aims to reduce ADC development timelines from 18 months to 12 months between clinical candidate nomination and Phase 1 trials, addressing the growing demand from over 180 companies developing more than 1,170 ADC candidates.
The collaboration provides an integrated solution for ADC manufacturing challenges, offering faster development processes while maintaining sustainability standards through green chemistry principles and waste reduction initiatives.
Italian pharmaceutical leader Indena and Princeton-based TCG GreenChem (搜索) have formed a strategic partnership to accelerate the development of next-generation payload-linkers (搜索) for antibody-drug conjugates (搜索) (ADCs (搜索)), addressing critical manufacturing bottlenecks in one of the fastest-growing segments of pharmaceutical innovation.
The collaboration combines Indena's three decades of expertise in high-potency active pharmaceutical ingredients (搜索) (HPAPI (搜索)) production with TCG GreenChem (搜索)'s advanced capabilities in green chemistry, catalysis, and biocatalysis. This partnership aims to provide ADC developers with faster, more efficient routes to clinical and commercial supply.
Addressing Growing Market Demand
The strategic alliance comes at a critical time for the ADC sector. As of mid-2025, over 180 companies are actively developing more than 1,170 ADC candidates, creating unprecedented demand for specialized manufacturing capabilities. This rapid growth places significant pressure on development and manufacturing infrastructure, requiring greater capacity and faster timelines while maintaining the highest quality standards.
"Partnering with TCG GreenChem (搜索) means working with an authoritative partner to address the needs of ADCs (搜索) developers," said Daniele Giavini, Managing Director at Indena. "Our expertise in high containment processes, designed to produce innovative HPAPIs, complements their deep capability in complex chemistry and process intensification, managed with a green approach."
Accelerated Development Timelines
The partnership promises to deliver substantial improvements in development efficiency. According to Gopal Sirasani, vice-president and head of business development at TCG GreenChem (搜索), "This collaboration could decrease the traditional 18-month timeline from clinical candidate nomination to Phase 1 trials down to just 12 months, saving a critical third of development time."
This acceleration is achieved through TCG GreenChem (搜索)'s "First-Time-Right with a Line of Sight" methodologies, which ensure precision from the outset. The company's team of 85 staff, including 60 PhD holders, brings expertise in catalytic chemistry, flow chemistry, and crystal engineering to drive innovation and accelerate development timelines.
Integrated Manufacturing Solutions
Traditional ADC manufacturing requires pharmaceutical companies to work with multiple partners handling different aspects, from payload synthesis to linker technology. The Indena-TCG GreenChem (搜索) partnership addresses this fragmentation by providing a cohesive, integrated service capable of handling both potent and non-potent compound challenges within a unified framework.
"We are experts in linker technology and non-potent chemistry. Our collaboration with Indena allows us to bridge the gap, delivering end-to-end solutions," explained Chris Senanayake, CEO of TCG GreenChem (搜索). "This means we can handle complex development challenges by harnessing Indena's HPAPI (搜索) expertise alongside our own strengths."
Bernard Vianes, global director of CDMO services at Indena, highlighted the technological advantages: "TCG has access to very innovative technology. Flow chemistry can be really useful for making linkers, bringing a new approach to their manufacture. There's a huge need for very specialised companies capable of this level of innovation."
Sustainability Focus
The partnership emphasizes environmental responsibility through green chemistry principles. TCG GreenChem (搜索)'s approach incorporates solvent minimization, waste reduction, and energy-efficient reactions. Indena's comprehensive sustainability program aligns with the United Nations' 2030 Agenda for Sustainable Development Goals, addressing sustainable sourcing, renewable energy utilization, and circular economy principles.
"We will drive a new era of greener, faster, and more flexible pharmaceutical development, aligned with the industry's evolving expectations," stated Senanayake.
Market Access and Commercial Benefits
The strategic alliance enhances both companies' presence in Europe and the US, two critical markets for pharmaceutical innovation and manufacturing. Joe Armstrong, Chief Marketing Officer of TCG GreenChem (搜索), noted that "for pharmaceutical clients, this means simplified supply chains, reduced costs, and an accelerated pathway from laboratory innovation to market-ready medicines."
The first milestone of the cooperation focuses on a novel payload-linker designed to showcase how this integrated approach can reduce development timelines and improve delivery to patients. The partnership is positioned to set new standards for pharmaceutical manufacturing that prioritize speed, sustainability, and precision in ADC development.
