The company's shares jumped from $0.27 to $1.32 at market close on October 30, bringing its market capitalization to $64.8 million.
Strong Disease Control Across Multiple Cancer Types
In the Phase I/II trial (NCT03058289), INT230-6 achieved a disease control rate of 75% in adult patients with metastatic disease across more than 20 cancer types, including pancreatic, breast, and sarcoma. The median overall survival reached 11.9 months, substantially exceeding the historical median overall survival of four to seven months for these patient populations.
The results were particularly striking in metastatic sarcoma patients, where median overall survival extended to 21.3 months. This represents a significant improvement over standard care outcomes in this difficult-to-treat cancer type.
Dose-Response Relationship Reveals Optimal Treatment Strategy
Exploratory analysis revealed a clear dose-response relationship based on tumor burden coverage. Patients treated at doses covering greater than 40% of total tumor burden achieved an 83.3% disease control rate compared to 50% in patients receiving lower doses. The survival benefit was even more pronounced, with median overall survival reaching 18.7 months in the higher-dose group versus just 3.1 months in the lower-dose cohort.
Rare Abscopal Effects Observed
Dr. Anthony El-Khoueiry, clinical research associate director at USC Norris (搜索), highlighted the therapy's unique mechanism of action. "Uninjected tumours shrinking from a locally administered therapy, referred to as abscopal effects, are generally rare for local therapies. Yet, an abscopal effect was observed in at least 20% of 48 patients who received drug volumes above 40% of their tumour burden," he stated.
The immune activation was further confirmed through biopsy analysis. "In 13 of 14 matched pair biopsy slides, a notable increase in activated CD4+ (搜索) and CD8+ (搜索) T cells was observed in the tumour microenvironment in response to INT230-6 treatment," El-Khoueiry noted. "The abscopal effects and immune cell infiltration observed in this study highlight this intertumoral therapy's potential to drive both a local and systemic anti-cancer activity."
Favorable Safety Profile
The treatment demonstrated a manageable safety profile with no dose-limiting toxicities reported. Seven patients experienced Grade 3 treatment-related adverse events, while no patients suffered Grade 4 or 5 treatment-related adverse events, indicating the therapy's tolerability across the diverse patient population.
Phase III Development Underway
Based on these encouraging results, Intensity Therapeutics has already initiated Phase III development. The INVINCIBLE-3 trial in sarcoma (NCT06263231) is currently underway, and the company has announced plans for a Phase III study in triple-negative breast cancer.
INT230-6 represents a novel approach to cancer treatment as a fixed-dose combination of two established chemotherapies, cisplatin and vinblastine sulphate. The therapy works by interfering with mitosis to inhibit DNA synthesis, delivering cytotoxic effects directly to the tumor microenvironment while potentially triggering systemic immune responses.
