Istesso Begins Phase II Trial of Leramistat for Sarcopenia in Rheumatoid Arthritis Patients

Istesso (搜索), a portfolio company of IP Group PLC (搜索), has begun dosing patients in a Phase II clinical trial of its lead drug candidate leramistat for the treatment of sarcopenia (搜索) caused by rheumatoid arthritis (搜索). The study, designated IST-03, represents a significant step forward for a condition that currently has no specific pharmacological therapies available.
The randomized, double-blind, placebo-controlled trial is designed to evaluate leramistat's effects on muscle quality, repair, and function. Enrollment is underway at Newcastle University and Newcastle Hospitals, in collaboration with the National Institute for Health and Care Research, whose advanced imaging and biopsy capabilities are well suited to the trial's requirements.
A Growing Unmet Need
Sarcopenia (搜索), characterized by progressive loss of muscle mass and function, represents a significant and growing unmet medical need across aging populations and chronic diseases. Despite its substantial burden on patients and healthcare systems, no specific pharmacological therapies are currently approved for the condition.
The IST-03 trial builds on earlier clinical data from rheumatoid arthritis (搜索) studies, in which leramistat demonstrated improvements in disability and fatigue while reducing markers of muscle loss. Preclinical investigations further showed that the drug restored muscle quality in disease models and recovered muscle mass and function in aged animals beyond levels observed in younger baseline animals.
A Novel Regenerative Mechanism
Leramistat is the lead candidate from Istesso (搜索)'s proprietary class of Mitochondrial Complex I (搜索) Modulators. These compounds are designed to trigger the body's own tissue repair and regeneration processes, a mechanism that has shown effects across multiple tissue types, including muscle, bone, gut, and lung.
Greg Smith, chief executive of IP Group, described the trial as an important inflection point for Istesso (搜索) as it advances its regenerative medicine approach into Phase II evaluation. "The programme is backed by a consistent body of clinical and preclinical data," Smith said, adding that "the ability to influence underlying tissue repair represents a differentiated and scalable opportunity."
Smith further noted that success in the IST-03 trial would validate the approach and open up opportunities across large areas of unmet medical need. IP Group holds an undiluted 56.5% stake in Istesso (搜索).
