Jade Biosciences Advances Three Monoclonal Antibody Programs for Autoimmune Diseases with Phase 1 Data Expected in 2026
核心洞察
Jade Biosciences (搜索) expects Phase 1 interim results for JADE101 (搜索), a novel anti-APRIL (搜索) monoclonal antibody for IgA nephropathy (搜索), in the first half of 2026 with Phase 2 initiation planned for mid-2026.
The company plans to begin first-in-human studies for JADE201 (搜索), a half-life extended anti-BAFF receptor antibody for rheumatoid arthritis (搜索), in the second quarter of 2026.
With approximately $336 million in cash as of December 31, 2025, Jade expects funding runway into the first half of 2028 to support its pipeline of potentially best-in-class autoimmune therapies.
Jade Biosciences (搜索) announced its 2026 strategic priorities, outlining key milestones for three monoclonal antibody programs targeting autoimmune diseases (搜索). The clinical-stage biotechnology company expects to report Phase 1 interim data for its lead candidate JADE101 (搜索) in the first half of 2026, followed by Phase 2 initiation for IgA nephropathy (搜索) patients mid-year.
JADE101 Targets IgA Nephropathy with Novel Anti-APRIL Approach
JADE101 (搜索) is an investigational, fully human monoclonal antibody designed to selectively inhibit A Proliferation-Inducing Ligand (APRIL (搜索)), a key driver of pathogenic IgA production in patients with IgA nephropathy (搜索) (IgAN). The progressive autoimmune kidney disease can lead to kidney failure (搜索), representing a significant unmet medical need.
Currently in Phase 1 testing in healthy volunteers, JADE101 (搜索) has demonstrated ultra-high binding affinity in preclinical studies and is engineered for half-life extension. The company aims to combine disease-modifying efficacy through a long dosing interval with patient-friendly subcutaneous dosing.
"The company believes selective anti-APRIL (搜索) therapies are poised to become foundational treatments in IgAN, potentially moving to frontline therapy by delivering disease-modifying efficacy, including durable reductions in pathogenic IgA and proteinuria and stabilization of kidney function, without unnecessary immune suppression," according to the company's announcement.
Phase 1 interim results from the ongoing healthy volunteer study expected in the first half of 2026 are anticipated to define dose and dose interval selection for Phase 2 and Phase 3 IgAN patient studies. Jade expects to initiate a Phase 2 clinical trial in patients with IgAN mid-2026, with preliminary data anticipated in 2027.
JADE201 Advances Enhanced Anti-BAFF Receptor Strategy
JADE201 (搜索) represents a half-life extended, enhanced effector function monoclonal antibody targeting the B-cell activating factor receptor (搜索) (BAFF-R (搜索)). The antibody is designed to build on the differentiated dual mechanism of prior anti-BAFF-R antibodies to deplete B cells while addressing limitations such as short half-life through the incorporation of half-life extension technology.
A first-in-human study initially evaluating JADE201 (搜索) in patients with rheumatoid arthritis (搜索) is expected to begin in the second quarter of 2026, with interim data anticipated in 2027. The randomized, placebo-controlled, single ascending dose trial will evaluate safety, tolerability, pharmacokinetics, and pharmacodynamics, incorporating biomarker-rich endpoints such as BAFF-R (搜索) occupancy, soluble BAFF levels, and B-cell subpopulation profiling.
Pipeline Expansion with Third Development Candidate
The company has nominated JADE301 (搜索) as its third development candidate, an undisclosed antibody program currently in preclinical testing. JADE301 is expected to enter first-in-human study in the first half of 2027, with further details expected to be disclosed in the second half of 2026.
Strong Financial Position Supports Development Timeline
Jade expects to report that its cash, cash equivalents and investments as of December 31, 2025 were approximately $336 million, which is expected to fund operations into the first half of 2028. This financial runway provides support for the company's planned clinical milestones across its three-program pipeline.
"Backed by a strong balance sheet with runway expected to last into the first half of 2028, we enter the year poised to deliver meaningful benefit to patients, have impactful clinical catalysts, and drive long-term growth and value creation," said Tom Frohlich, Chief Executive Officer of Jade Biosciences (搜索).
The company will present its 2026 outlook at the 44th Annual J.P. Morgan Healthcare Conference on Thursday, January 15, 2026, at 11:15 a.m. PT, where management will also host one-on-one investor meetings during the conference.
