Approximately 80% of affected individuals experience disease onset during infancy, and many of these children never achieve fundamental developmental milestones such as walking or speaking. Some do not survive into adulthood. For those with later-onset disease, the condition typically manifests as disruptive nocturnal seizures accompanied by significant cognitive and psychiatric comorbidities.
ABS-1230 is designed as a potent and selective orally available small molecule KCNT1 (搜索) ion channel inhibitor. In preclinical studies, the compound inhibited KCNT1 across all evaluable pathogenic mutations, suggesting potential to treat all patients with KCNT1-related epilepsy. In an early clinical proof-of-concept trial, children with KCNT1-related epilepsy who received ABS-1230 experienced meaningful seizure reductions.
Regulatory Momentum and the KYRON Trial
ABS-1230 has secured multiple FDA designations that underscore both the severity of the unmet need and the agency's interest in facilitating rapid development: Fast Track, Rare Pediatric Disease, and Orphan Drug Product designations. Notably, the therapy has also been accepted into the FDA's new Rare Disease Evidence Principles (RDEP) program, which aims to accelerate the development of ultra-rare disease therapies.
The ongoing Phase 1b/2a KYRON trial is designed to serve as the registrational study to support a new drug application submission in the United States.

The new company, funded by existing investors, will focus on genetic rare neurological diseases, including a clinical-stage small molecule TRPV4 (搜索) inhibitor, ABS-0871, for Charcot-Marie-Tooth type 2C (搜索) and other early-stage programs. Jazz will receive a minority stake and certain related rights in the new entity.
