KaliVir Partners with Roche to Test Novel Oncolytic Immunotherapy VET3-TGI in Combination with Tecentriq
核心洞察
KaliVir Immunotherapeutics (搜索) announced a clinical trial collaboration with Roche to evaluate VET3-TGI combined with atezolizumab (Tecentriq) in advanced solid tumors (搜索).
The partnership will expand KaliVir's ongoing STEALTH-001 Phase 1a/1b study, with Roche supplying atezolizumab for multiple dosing cohorts.
VET3-TGI is designed to destroy tumor cells while activating immune responses through IL-12 (搜索) and TGF-β (搜索) inhibitor expression.
KaliVir Immunotherapeutics (搜索) has entered into a clinical trial collaboration and supply agreement with Roche to evaluate its lead oncolytic immunotherapy candidate VET3-TGI in combination with Roche's checkpoint inhibitor atezolizumab (Tecentriq) for patients with advanced solid tumors (搜索).
The collaboration will support the expansion of KaliVir's ongoing STEALTH-001 Phase 1a/1b clinical study, which is currently evaluating VET3-TGI in patients with advanced or metastatic solid tumors (搜索). Under the agreement terms, Roche will supply atezolizumab for use in combination with VET3-TGI across multiple dosing cohorts.
Novel Oncolytic Platform Technology
VET3-TGI represents KaliVir's lead therapeutic candidate, developed using the company's proprietary Vaccinia Enhanced Template (VET™) platform. The oncolytic virus is designed to selectively replicate in tumor cells while simultaneously activating potent immune responses through a proprietary payload that includes interleukin-12 (搜索) (IL-12 (搜索)) and a TGF-β (搜索) inhibitor.
The combination of VET3-TGI with atezolizumab aims to amplify immune activation and reshape the tumor microenvironment, with the goal of broadening and deepening clinical benefit for patients across a range of solid tumors.
"This clinical collaboration with Roche allows us to build on the promising clinical and preclinical data by expanding the evaluation of VET3-TGI beyond monotherapy into combination therapy with an established checkpoint inhibitor," said Helena Chaye, CEO of KaliVir Immunotherapeutics (搜索). "Partnering with Roche, a global leader in oncology, represents an important step toward advancing innovative therapeutic strategies and exploring the full potential of this novel combination for patients with advanced solid tumors (搜索)."
STEALTH-001 Study Design
The STEALTH-001 study (NCT06444815) is a first-in-human, open-label, dose escalation and expansion Phase 1a/1b trial assessing both intratumoral and intravenous administration of VET3-TGI. The trial evaluates VET3-TGI as both a monotherapy and in combination with a checkpoint inhibitor in patients with pathologically confirmed, advanced, unresectable or metastatic solid tumors (搜索).
The study continues to progress as planned through its dose escalation phase, with the trial designed to evaluate VET3-TGI administered through direct intratumoral injection and intravenous infusion.
Platform Technology Advantages
KaliVir's proprietary VET™ platform integrates multiple genetic modifications, allowing for the systemic delivery of oncolytic vaccinia candidates and the targeted expression of therapeutic transgenes within tumors. The platform harnesses the unique advantages of the vaccinia virus backbone, which the company engineers to create optimized viral candidates for cancer (搜索) treatment.
The oncolytic virus is designed to selectively replicate in tumor cells, stimulate local immune responses, and remodel the immunosuppressive tumor microenvironment through the expression of IL-12 (搜索) and a TGF-β (搜索) inhibitor, potentially offering a multi-mechanistic approach to cancer (搜索) immunotherapy.
