The biotechnology startup, backed by investors including Fairmount, Venrock, Wellington Management, and TCGX, is developing what it positions as a next-generation approach to clearing amyloid beta (搜索) plaques in the brain.
Targeting Blood-Brain Barrier Challenges
KRSA-028 (搜索) represents an attempt to overcome one of the most significant obstacles in neurodegenerative drug development: the blood-brain barrier. According to the company, very little of traditional monoclonal antibodies makes it past this barrier, reducing efficacy and requiring higher dosages that can cause safety issues.
Korsana is leveraging specialized technology to shuttle its drug through the blood-brain barrier using less frequent subcutaneous administration. This approach contrasts with current approved treatments, where Eli Lilly's Kisunla is only available as an intravenous infusion, while patients on Eisai and Biogen's Leqembi can progress to weekly subcutaneous administration after 18 months of intravenous infusions.
Addressing Current Treatment Limitations
"Only two disease-modifying therapies have been approved to treat Alzheimer's, and both carry safety warnings, offer only modest efficacy, and impose a high burden of care," said Korsana president and CEO Jonathan Violin in a statement. "Patients deserve better options than what is currently available."
The company is betting that KRSA-028 (搜索) can help clear amyloid plaque like existing approved treatments while sidestepping adverse side effects such as brain swelling and micro-bleeds that have been associated with current therapies.
Development Timeline and Corporate Structure
Korsana plans to have proof of concept data for KRSA-028 (搜索) by the end of 2027. The drug was originally developed by Paragon Therapeutics (搜索), a hub-and-spoke biotech that launches spinouts focused on single assets. Paragon's roster of previous "spoke" companies includes Apogee Therapeutics and Crescent Biopharma.
Beyond its lead program, Korsana has two other unnamed programs in development for neurodegenerative conditions, suggesting a broader pipeline approach to addressing unmet medical needs in this therapeutic area.
Funding and Leadership
The company was first established with a $25 million seed round in 2024, backed by Fairmount and Venrock Healthcare Capital Partners. The Series A round, co-led by Wellington Management and TCGX in September, brought in an additional $150 million from investors including J.P. Morgan Life Sciences Private Capital, Janus Henderson, Sanofi Ventures, and Foresite Capital.
Korsana is led by Jonathan Violin, a partner at Fairmount and former CEO of Viridian Therapeutics, bringing experience in biotech leadership to the emerging company's mission to develop improved Alzheimer's treatments.
