Clinical Trial Results Show Promise
Clinical trials conducted by doctors at Zealand University Hospital in Roskilde have yielded encouraging preliminary results. In double-blind studies, patients with mild to moderate Alzheimer's disease (搜索) spent 30 minutes daily over periods of either six or 12 weeks in front of specialized light devices or placebo lamps. Neither patients nor doctors knew which type of treatment was being administered.
The results demonstrated measurable improvements in cognitive abilities, particularly in speech and memory functions, among patients who received the active light treatment. Brain scans provided additional evidence of the therapy's effectiveness, revealing structural improvements in treated patients.
"In the patients who were given the placebo lamp, that part of the brain deteriorated, while we actually saw a small increase in volume in those who used an active lamp," explained DTU (搜索) Professor Paul Michael Petersen, one of the technology's inventors.
Mechanism and Scientific Foundation
The therapy works by inducing specific gamma brain waves (搜索) that appear to benefit Alzheimer's patients. The concept builds upon research from MIT conducted nearly a decade ago, which showed that exposing mice with Alzheimer's to blinking lights induced gamma brain waves and reduced harmful beta-amyloid (搜索) protein plaques in the brain.
However, traditional blinking lights pose significant risks, including discomfort and potential epileptic seizures. The DTU (搜索)-Berkeley team addressed these concerns by developing a device that achieves the same beneficial brain effects without users detecting the light's fluctuation.
"Blinking lights are not particularly sound because they can cause discomfort and, in the worst case, epileptic seizures, so we wanted to investigate whether we could achieve the same effect but in a gentler way," Petersen noted.
Technology Development and Validation
The device underwent rigorous testing, beginning with healthy individuals to confirm its ability to induce the desired brain waves. The technology was initially developed during a three-week course by Marcus Carstensen, then a master's student at DTU (搜索), who created the first electrical circuit using new LED technology. Carstensen, now Chief Technology Officer at OptoCeutics (搜索), continued developing the device through his master's project and PhD studies.
The Danish-American partnership originated in 2017 when DTU (搜索) Professor Jes Broeng established connections between brain researchers at UC Berkeley (搜索) and light experts at DTU during his stay at the California institution.
Future Applications and Ongoing Research
OptoCeutics (搜索) is currently conducting a larger study to provide stronger clinical evidence for the technology's effectiveness. The devices are already available on the market, and successful completion of the expanded trials could lead to broader adoption in hospitals and clinics treating Alzheimer's disease (搜索).
Long-term vision for the technology extends beyond clinical settings. Petersen envisions developing the concept into preventive lighting solutions for home use, with particular relevance for retirement homes where dementia (搜索) prevalence is high.
"If we can stimulate patients' brains so they avoid secondary diseases and see an improvement in their daily lives by reducing the consequences of losing their memory and not being able to orient themselves or speak, then that's huge!" Petersen emphasized.
Disease Context and Unmet Need
Alzheimer's disease (搜索) represents the most common form of dementia (搜索) and ranks as the fourth leading cause of death in Denmark. The Danish Dementia Research Centre estimates approximately 25,000 elderly people in Denmark are diagnosed with Alzheimer's disease. Current treatment options remain limited, with available drugs providing only temporary stabilizing effects on disease progression. In most cases, the disease occurs without known hereditary factors or identifiable causes.
