Addressing Unmet Medical Needs
The patent covers administration of therapeutically effective amounts of isolated allogeneic mesenchymal stem cells (搜索) (MSCs) to patients with aging-related frailty (搜索) with inflammaging and those with non-ischemic dilated cardiomyopathy (搜索). Aging-related frailty is a syndrome of multisystem physiological dysregulation occurring in older individuals which diminishes human healthspan. The condition is characterized by weakness, low physical activity, slowed motor performance, exhaustion, and unintentional weight loss, with no approved treatments currently available.
Many studies show a direct correlation between aging-related frailty (搜索) and inflammation. Inflammaging has received considerable attention because it proposes a link between immune changes and a number of diseases and conditions common in old age, including aging-related frailty.
Clinical Development Progress
Longeveron has previously conducted Phase 1 and 2 studies in aging-related frailty (搜索) designed to show improvements in the six-minute walk test and physical functioning for laromestrocel. The mesenchymal stem cell therapy has delivered positive initial results across two clinical trials in aging-related frailty.
Cardiac Applications
Non-ischemic dilated cardiomyopathy (搜索) (NIDCM) is a condition where the heart muscle becomes weakened and enlarged, negatively affecting its ability to pump blood effectively. NIDCM is a progressive disorder with no current cure, often leading to heart transplantation. In vivo administration of isolated MSCs has been shown to favorably alter several immunologic markers typically elevated in chronic inflammation. Restoration of immune competence has clinical relevance in patients who are of higher risk for co-morbid infectious disease.
Therapeutic Mechanism
Laromestrocel is a living cell product made from specialized cells isolated from the bone marrow of young healthy adult donors. These mesenchymal stem cells (搜索) are essential to the endogenous biological repair mechanism and have been shown to perform complex functions in the body, including the formation of new tissue. They also respond to sites of injury or disease and secrete bioactive factors that are immunomodulatory and regenerative.
The company believes that laromestrocel may have multiple potential mechanisms of action that may lead to anti-inflammatory, pro-vascular regenerative responses, and therefore may have broad application for a range of rare and aging related diseases.
Current Pipeline
Longeveron is currently pursuing three pipeline indications: hypoplastic left heart syndrome (搜索) (HLHS), Alzheimer's disease (搜索), and Pediatric Dilated Cardiomyopathy (搜索) (DCM). Laromestrocel development programs have received five distinct FDA designations: for the HLHS program - Orphan Drug designation, Fast Track designation, and Rare Pediatric Disease designation; and, for the AD program - Regenerative Medicine Advanced Therapy (RMAT) designation and Fast Track designation.
