Major Cancer Research Coalition Launches $12 Million Initiative to Advance Early Detection of Deadly Cancers

The Mark Foundation for Cancer Research (搜索) has announced the formation of an unprecedented coalition with leading cancer research organizations to tackle one of oncology's most pressing challenges: early detection of the world's deadliest cancers. The coalition, comprising the American Association for Cancer Research (搜索) (AACR), Lustgarten Foundation (搜索), Break Through Cancer (搜索), and The Honorable Tina Brozman Foundation (Tina's Wish (搜索)), will invest $12 million across six collaborative research projects targeting pancreatic, ovarian, and esophageal cancers, as well as cancer predisposition syndromes.
Critical Need for Early Detection Breakthrough
These cancer types present a formidable clinical challenge, often growing and spreading silently in their early stages, which contributes to persistently poor long-term survival rates. Current screening technologies have largely failed to detect these malignancies early enough to meaningfully improve patient outcomes. Pancreatic cancer (搜索) exemplifies this challenge with a five-year survival rate of just 13%, largely attributed to late detection, according to Linda Tantawi, CEO of the Lustgarten Foundation (搜索).
"We can't cure what we can't detect, and late-stage diagnosis often limits treatment options," said Ryan Schoenfeld, PhD, CEO of The Mark Foundation. "For too long, significant advances in early detection of these deadly cancer types have been lacking. This coalition breaks down barriers between funders, scientists, and research disciplines to drive faster progress."
The situation is equally dire for ovarian cancer (搜索), where no effective screening test currently exists. Amy Kyle, Board Chair of Tina's Wish (搜索), noted that "even cancers caught at a so-called early stage are incredibly difficult to eradicate."
Innovative Research Approaches
The coalition has awarded up to $2 million to each of six research teams pursuing groundbreaking approaches to overcome the most pressing obstacles in early cancer detection:
KRAS-Targeted Detection Strategies
Two projects focus on KRAS (搜索) mutations, which are prevalent in pancreatic cancer (搜索). Nikolaos G. Sgourakis, PhD, from Children's Hospital of Pennsylvania and University of Pennsylvania, leads a team developing programmable recognition of KRAS neoantigens for early cancer diagnostics across patients. A complementary project led by Brian M. Wolpin, MD, MPH, and Andrew J. Aguirre, MD, PhD, from Dana-Farber Cancer Institute aims to detect and intercept KRAS-mutant pancreatic cancer using small molecule RAS(ON) inhibitors (搜索).
Advanced Genomic and Digital Pathology Methods
The research portfolio includes sophisticated approaches leveraging cutting-edge technologies. Dan Landau, MD, PhD, from Weill Cornell Medicine leads a project harnessing extrachromosomal DNA (ecDNA) and aneuploidy signals in plasma whole genome sequencing for early detection of ovarian cancer (搜索). Christina Curtis, PhD, and Greg Charville, MD, PhD, from Stanford University are developing digital pathology diagnostics for robust stratification of esophageal cancer (搜索) risk.
Molecular Profiling and Predisposition Syndromes
Peter K. Sorger, PhD, and colleagues at Harvard University are conducting molecular profiling of ovarian cancer (搜索) precursors to transform early detection and precancer stratification. Additionally, Trevor J. Pugh, PhD, from Princess Margaret Cancer Centre is working toward a unified platform for Li-Fraumeni syndrome (搜索) cancer risk prediction and cell-free DNA surveillance.
Collaborative Model for Accelerated Progress
This initiative represents an ambitious shift in cancer research funding by aligning strategy and uniting expertise to accelerate solutions that no single organization could achieve alone. The coalition emerged from a scientific workshop organized in Philadelphia, Pennsylvania, last January by three of the partners, which brought together dozens of scientists and physician-scientists to forge the partnerships now being formalized through the grant program.
"Knowledge-sharing is central to solving the most pressing challenges in cancer research," said Dr. Margaret Foti, PhD, MD (hc), CEO of AACR. "By leveraging our collective expertise, we have brought together a network of international experts across various scientific disciplines to address cancers that have historically been very difficult to detect."
Tyler Jacks, PhD, President of Break Through Cancer (搜索), emphasized the potential of emerging technologies: "New cutting-edge technologies such as artificial intelligence and single-cell profiling are providing new opportunities to decipher hidden early disease biology, predict patient outcomes, and accelerate translational progress."

Beyond providing funding, the initiative is designed to build a global early detection ecosystem that could transform cancer care paradigms. The coalition's approach pools $12 million in joint investment while aligning the expertise of top scientists across disciplines to overcome scientific hurdles that have long impeded progress in early cancer detection.
The Mark Foundation for Cancer Research (搜索), which has awarded more than $300 million in grants to investigators at more than 120 academic institutions across 18 countries since 2017, continues to expand its impact through strategic partnerships and collaborative funding models that address the most challenging aspects of cancer research and treatment development.
