"Our study describes the events that occur when a patient's tumor harbors only one copy of a cancer (搜索)-causing gene, which occurs in 20% of cases," explains Alan P. Fields, Ph.D., cancer biologist at Mayo Clinic Comprehensive Cancer Center (搜索) and principal investigator of the study.
The research team found that the missing PRKCI (搜索) gene results in tumors that grow less aggressively compared to those with intact gene copies. Surprisingly, these tumors still develop despite lacking a gene typically essential for tumor proliferation, prompting graduate student Joey Nguyen to investigate this paradox.
Early experiments revealed that when PRKCI (搜索) is missing, lung adenocarcinoma (搜索) cells acquire characteristics of lung cells that regenerate tissue after damage. Collaborating with systems biology researcher Hu Li, Ph.D., the team examined this process at the single-cell level.

The findings identify three tumor characteristics that may help clinicians identify optimal candidates for immune checkpoint inhibitors (搜索):

"Now that we understand how PRKCI (搜索) is working in a lung tumor, it may be possible to couple a PRKCI inhibitor (搜索) with immunotherapy (搜索), so a future clinical trial that combines these approaches will certainly be an important avenue to explore," Fields concludes.
