Moderna Advances mRNA Hantavirus Vaccine Development as Deadly Cruise Ship Outbreak Highlights Urgent Medical Need
核心洞察
Moderna has partnered with Korea University College of Medicine (搜索) since 2023 to develop an experimental mRNA-based hantavirus (搜索) vaccine targeting multiple strains of the deadly virus.
The U.S. Army has completed Phase 1 clinical trials for DNA vaccines against three hantavirus (搜索) strains, including the Andes virus (搜索) responsible for the recent cruise ship outbreak.
The Andes virus (搜索) strain has a fatality rate of 30-50% and is the only hantavirus (搜索) known to spread between humans, making vaccine development critically important.
A deadly hantavirus (搜索) outbreak aboard the cruise ship MV Hondius has intensified global efforts to develop vaccines against one of the world's most lethal rodent-borne diseases, with Moderna emerging as a key player through its experimental mRNA-based vaccine program. The outbreak, caused by the Andes virus (搜索) strain, has killed three passengers and infected several others, highlighting the urgent need for effective preventive measures against a pathogen with fatality rates reaching up to 50%.
Moderna's mRNA Vaccine Initiative
In 2023, Moderna partnered with the Vaccine Innovation Center at Korea University College of Medicine (搜索) to develop an experimental mRNA-based hantavirus (搜索) vaccine under the company's 'mRNA Access' program. The collaboration, which became public in 2024, focuses on creating broader protection against multiple hantavirus strains rather than targeting only one variant.
"Current vaccines used in parts of Asia offer incomplete coverage and are largely designed for older hantavirus (搜索) strains associated with hemorrhagic fever and kidney disease," according to researchers involved in the project. The latest effort aims to use mRNA technology similar to Moderna's COVID-19 vaccine platform, allowing scientists to rapidly redesign shots against emerging and more dangerous strains.
U.S. Army's Decades-Long Research Program
The U.S. Army Medical Research Institute of Infectious Diseases (搜索) (USAMRIID) has been at the forefront of hantavirus (搜索) vaccine development for over three decades. Jay Hooper, Chief of the Molecular Virology Branch at USAMRIID, who has studied hantavirus for over 30 years, explained the military's interest: "Because rodent-borne Hantavirus is a direct threat to soldiers in the field, the military has long wanted a vaccine."
To date, Phase 1 clinical trials have been completed for three strains: Andes virus (搜索), Hantaan virus, and Puumala virus. The Andes virus DNA vaccine (搜索) works by injecting the virus's genetic information in DNA form to induce an immune response, generating neutralizing antibodies (搜索) that directly disable the virus in the human body.
"The outlook is promising," Hooper said, but noted a significant limitation: "it requires at least three doses." The team is currently verifying the vaccine's effectiveness by administering neutralizing antibodies (搜索) extracted from Phase 1 participants to hamster models, with results expected to be published in an upcoming paper.
Clinical Development Challenges
Despite promising Phase 1 results, advancing to Phase 2 and 3 trials presents unique challenges. "Because cases are rare and geographically dispersed, it is difficult to identify a specific region to conduct traditional efficacy trials," Hooper explained. "We need a creative approach that uses the sufficient generation of virus-neutralizing antibodies (搜索) after vaccination as a marker of effectiveness."
Recent laboratory findings published in Nature Communications in 2026 found that experimental nucleic-acid-based hantavirus (搜索) vaccines generated strong neutralizing antibody responses in pre-clinical testing, offering hope that modern vaccine platforms may finally overcome years of scientific setbacks.
Innovative Delivery Solutions
While vaccine efficacy remains a primary focus, researchers are also addressing delivery challenges. Professor Asel Sartbaeva of the University of Bath is developing innovative delivery methods through "ensilication" technology, which she has been developing for over 15 years.
"Ensilication is a groundbreaking new technology that enhances thermal stability by encasing the vaccine in a very thin layer of inorganic material," Sartbaeva explained. Her team is collaborating with a Texas research team and the South African biotech company Afrigen (搜索) with an ambitious goal: "We are aiming for a level that would even allow for drone delivery."
Therapeutic Development and Market Impact
Beyond vaccines, therapeutic development is also progressing. An antibody product called 'SAB-1634 (搜索),' created by injecting hantavirus (搜索) antigens into genetically modified cattle that produce human antibodies, has shown effectiveness against the Andes virus (搜索) and three other strains in animal models, though it has not yet entered Phase 1 human clinical trials.
The recent cruise ship outbreak has also influenced financial markets, with Moderna shares briefly surging after reports linked the company's vaccine research to the ongoing outbreak. However, analysts have warned that the disease remains a relatively small commercial market despite its high fatality rate.
Global Regulatory Landscape
Currently, no FDA-approved hantavirus (搜索) vaccine has reached widespread public rollout in Western countries. Chinese and Korean hantavirus vaccines exist but are not used in the US or Europe, primarily due to stricter regulatory standards, limited cross-protection against local virus strains, and uncertain effectiveness.
The Andes virus (搜索), responsible for the recent outbreak, is particularly concerning as it is the only hantavirus (搜索) known to spread between humans and has a case fatality rate frequently estimated between 30% and 50%. The virus causes Hantavirus Pulmonary Syndrome (搜索) (HPS), characterized by rapid-onset respiratory failure that can progress to acute respiratory distress syndrome (搜索) or multiple organ failure.
