Montai Therapeutics Advances First-in-Class Oral NRF2 Agonist for Ulcerative Colitis to IND-Enabling Studies
核心洞察
Montai Therapeutics (搜索) presented preclinical data on MTAI-1025 (搜索), a novel oral NRF2 (搜索) agonist that demonstrated superior efficacy compared to monoclonal antibody therapy in ulcerative colitis (搜索) mouse models.
The compound showed dose-dependent reduction in disease activity and weight loss while improving mucosal architecture and reducing inflammatory infiltration with favorable tolerability.
MTAI-1025 (搜索) represents the first development candidate from Montai's AI-driven CONECTA platform to advance to IND-enabling studies, targeting three key mechanisms of ulcerative colitis (搜索).
Montai Therapeutics (搜索) has announced promising preclinical results for MTAI-1025 (搜索), a novel oral NRF2 (搜索) agonist that demonstrated superior efficacy to existing monoclonal antibody therapy in ulcerative colitis (搜索) models. The company presented the data at the American College of Asthma, Allergy and Immunology annual meeting on November 8th in Orlando, marking the first public disclosure of results for their lead development candidate.
Novel Mechanism Targets Multiple UC Pathways
MTAI-1025 (搜索) represents a first-in-class oral therapy that activates the transcription factor NRF2 (搜索), described as a master regulator of cytoprotective gene networks. The compound addresses ulcerative colitis (搜索) through a comprehensive approach targeting three primary disease mechanisms: epithelial barrier dysfunction, chronic mucosal inflammation, and oxidative stress.
"These data demonstrate that MTAI-1025 (搜索) can effectively attenuate inflammation and promote mucosal protection in vivo in preclinical models, supporting its potential as a first-in-class oral NRF2 (搜索) therapy for UC, both as a monotherapy or in combination with monoclonal antibodies," said Lamya Shihabuddin, Ph.D., Montai's Chief Scientific Officer.
Superior Efficacy in Preclinical Models
In a 14-day colitis (搜索) mouse model study, MTAI-1025 (搜索) demonstrated clear dose-dependent improvements across multiple outcome measures. Mice treated with low, medium, and high oral daily doses of MTAI-1025 showed superior efficacy compared to an anti-IL-12p40 (搜索) monoclonal antibody administered at 10 mg/kg via intraperitoneal injection.
Key findings included dose-dependent reductions in the UC Disease Activity Index and weight loss, with MTAI-1025 (搜索) outperforming the monoclonal antibody comparator. Histological analyses revealed improved mucosal architecture and reduced inflammatory infiltration, accompanied by marked reductions in pro-inflammatory cytokines and confirmed NRF2 (搜索) activation. The compound maintained a favorable tolerability profile with no signs of increased inflammation or necrosis.
AI-Driven Drug Discovery Platform
MTAI-1025 (搜索) emerged from Montai's CONECTA platform, which utilizes artificial intelligence to decode diverse chemistry from the company's Anthromolecule collection of over 1 billion bioactive molecules. These molecules are derived from compounds humans have chronically consumed in food, supplements, and herbal medicines, providing access to vast structural diversity with potential for selective biological modulation.
The compound represents the first development candidate produced by Montai's platform to advance to IND-enabling studies, demonstrating the potential of AI-driven approaches to address complex inflammatory diseases traditionally dominated by biologic therapies.
Addressing Unmet Medical Need
Ulcerative colitis (搜索), characterized as an autoimmune disease (搜索) of the gastrointestinal system, currently has limited effective treatment options. Existing biologic therapies do not fully address the complex interlinked mechanisms driving the disease. NRF2 (搜索) has shown promise in promoting antioxidant responses, suppressing pro-inflammatory signaling, and supporting epithelial repair in other immune-mediated diseases, but has previously eluded selective targeting with small molecules.
Shihabuddin emphasized the broader implications: "This is also an important demonstration of the power of Montai's platform to deliver potential oral medicines for complex, chronic inflammatory diseases that today are the exclusive domain of biologics."
Based on these preclinical results, Montai plans to advance MTAI-1025 (搜索) into IND-enabling studies, representing a significant milestone for both the compound and the company's AI-driven drug discovery approach.
