The collaboration aims to address significant unmet medical needs in brain cancer treatment through advanced drug delivery technology.
Novel Combination Therapy Approach
GLIORA (搜索) combines olaparib (originally developed by AstraZeneca as Lynparza®) and temozolomide (originally developed by Merck & Company (搜索) as Temodar®) in a locally-administered, long-acting, thermo-responsive hydrogel formulation. The treatment is designed to overcome the limitations of current standard-of-care oral temozolomide through localized delivery directly to the tumor resection bed during surgery.
By incorporating olaparib, a poly (ADP-ribose) polymerase (搜索) (PARP (搜索)) inhibitor that is mechanistically synergistic with temozolomide, the formulation amplifies chemo and radiotherapy-induced DNA damage while suppressing tumor repair pathways. This approach is expected to enhance tumor control during the critical post-surgical window, reduce the likelihood of resistance and recurrence, and minimize systemic exposure and side effects.
Advanced Manufacturing Technology
Under the partnership agreement, Nanoform will utilize its proprietary Controlled Expansion of Supercritical Solutions (CESS®) technology under GMP conditions to nanoform the APIs and intermediates at its Helsinki facility. The company will also employ its proprietary thermo-responsive hydrogel technology to develop the finished product prototype.

Clinical Development Timeline
Revio Therapeutics (搜索) will lead the preclinical and clinical development of the program and will be responsible for eventual manufacturing and supply of the final sterile dosage form. Prototype development and testing is currently at an advanced stage, with the program expected to enter clinical trials by the second half of 2026.
Subject to successful co-development and commercialization, GLIORA (搜索) could be commercially available by 2029-30. While olaparib is not currently approved for glioblastoma multiforme (GBM), its oral co-administration with temozolomide has shown promise in clinical settings, despite dose-limiting side effects that the new formulation aims to address.
Partnership Structure and Financial Terms
Development costs and all licensing and commercial revenues will be shared equally between the partners, with Nanoform receiving an additional €1.5 million in accelerated revenue-share payments. This collaborative approach leverages the complementary expertise of both companies in advancing innovative cancer treatments.
