In the phase 3 SIMPLIFY-1 trial, 67% of patients treated with Ojjaara achieved transfusion independence by week 24, significantly outperforming the 49% rate observed with Jakafi (ruxolitinib).
Addressing Multiple Disease Hallmarks Simultaneously

Transfusion independence was defined as avoiding transfusions while maintaining hemoglobin levels above 8 g/dL for at least 12 weeks.
Significant Clinical Burden of Anemia in Myelofibrosis
The prevalence of anemia (搜索) in myelofibrosis (搜索) presents a substantial clinical challenge. According to Palandri's research, approximately 40% of patients present with hemoglobin levels below 10 g/dL at diagnosis, with this percentage increasing to around 60% within the first year. Additionally, about one-quarter of patients require transfusions at diagnosis, and this number nearly doubles during the first year of disease progression.
"Any degree of anemia (搜索) correlates with decreased survival — patients with severe anemia have a median survival that's four times shorter than those with no anemia at all," Palandri noted. This correlation has led to the inclusion of low hemoglobin as a key factor in all current prognostic scoring systems for myelofibrosis (搜索).
Economic and Quality of Life Impact
Recent analysis conducted by Palandri's team in Italy revealed the substantial economic burden associated with transfusion-dependent myelofibrosis (搜索). Patients requiring transfusions spent six times more time in hospitals compared to those without anemia (搜索), while direct healthcare costs were 40 times higher for the National Health System. Indirect costs, including lost productivity for both patients and caregivers, were six times greater in transfusion-dependent patients.
Beyond economic considerations, anemia (搜索) significantly impacts patient quality of life. "Transfusion dependency correlates with a worse quality of life and a greater symptom burden, especially fatigue, which is linked to reduced productivity and social engagement," Palandri explained. Patients reported significant disruption across all aspects of their lives, including social, work, and daily activities.
Survival Implications and Future Treatment Goals
The achievement of transfusion independence with Ojjaara has been linked to improved overall survival outcomes. "We are accumulating more and more evidence that directly correlates resolution of anemia (搜索) and achievement of transfusion independence with longer overall survival," Palandri stated. This correlation positions anemia resolution as a critical therapeutic target in myelofibrosis (搜索) management.
The ability to address all three hallmarks of myelofibrosis (搜索) simultaneously may lead to improved long-term outcomes. As Palandri noted, "Once we can target all three of these features together, this can result in an improvement in overall survival. I think this is going to become one of our major therapeutic targets in the future."
The results support making transfusion independence a central treatment goal, particularly for patients presenting with low hemoglobin levels and reduced platelet counts, representing a significant advancement in myelofibrosis (搜索) therapeutic approaches.
