OncoC4 Launches AdaMunix Subsidiary to Address Anti-Drug Antibody Challenges in Protein Therapeutics
核心洞察
OncoC4 (搜索) announced the formation of AdaMunix (搜索), a new subsidiary focused on eliminating anti-drug antibodies (ADAs) that diminish protein therapeutic efficacy and cause adverse events.
AdaMunix (搜索) has developed two complementary platforms: ADA-X technology that neutralizes ADAs when combined with approved drugs, and BCR-X technology that creates ADA-resistant protein therapeutics.
In vivo studies have demonstrated near-complete elimination of ADAs for several approved drugs, with three programs advancing to pre-IND development based on promising preclinical results.
OncoC4 (搜索), Inc., a late-stage biopharmaceutical company developing novel cancer (搜索) medicines, has announced the formation of AdaMunix (搜索), a new subsidiary dedicated to addressing the pervasive challenge of anti-drug antibodies (ADAs) in protein-based therapeutics. The company will present its breakthrough technologies at the 2025 Orphan Drug Summit in Boston on July 17, 2025.
Novel Platform Technologies Target ADA Elimination
AdaMunix (搜索) has developed two complementary platform technologies designed to overcome ADA-related therapeutic limitations. The ADA-X technology creates drug-specific agents that neutralize ADAs when combined with approved therapeutics, while the BCR-X technology enables development of next-generation ADA-stealth protein therapeutics inherently resistant to ADA activity.
"ADAs are a pervasive barrier to protein-based therapeutics, diminishing drug efficacy and contributing to significant adverse events," said Yang Liu, PhD, Co-Founder, Chief Executive Officer and Chief Scientific Officer of OncoC4 (搜索). "To overcome these challenges, we've established multiple platforms designed to eliminate ADAs and enhance the efficacy and safety of antibody, enzyme, and gene therapies."
Promising Preclinical Results Drive Program Advancement
In vivo studies have demonstrated near-complete elimination of ADAs for several approved drugs using AdaMunix (搜索)'s platform technologies. Based on these promising preclinical results, the subsidiary has advanced three programs to pre-IND development stage, representing significant progress toward clinical evaluation.
The multipronged approach aims to harness these technologies to make a dramatic impact on patient lives across a wide range of therapeutic indications, according to Liu. The platforms are designed to enhance the efficacy and safety of antibody, enzyme, and gene therapies by addressing the fundamental challenge of immunogenicity.
OncoC4's Broader Pipeline and Strategic Collaborations
OncoC4 (搜索), based in Rockville, Maryland, maintains an active pipeline featuring assets with first-in-class and best-in-class potential targeting both novel and well-validated targets across oncology and immunological diseases. The company's wholly-owned AI-081, a potentially best-in-class bispecific antibody candidate targeting PD-1 (搜索) and VEGF (搜索), is currently enrolling patients in a Phase 1/2 trial that could enable two or more pivotal studies.
Additionally, OncoC4 (搜索) has established a strategic collaboration with BioNTech to co-develop gotistobart (BNT316/ONC-392), a next-generation anti-CTLA-4 (搜索) antibody candidate, in multiple solid tumor indications. This collaboration includes an ongoing pivotal clinical trial in squamous non-small cell lung cancer (搜索).
The formation of AdaMunix (搜索) represents OncoC4 (搜索)'s commitment to addressing fundamental challenges in protein therapeutics development, potentially transforming treatment outcomes for patients requiring these critical medicines.
