The company has received IND clearance from the U.S. Food and Drug Administration and plans to initiate a Phase 2 clinical trial in IPF patients in 2026.
Novel Mechanism Targets Root Cause of Fibrosis
ORJ-001 (搜索) represents a paradigm shift in IPF (搜索) treatment by targeting alveolar epithelial type 2 (AEC2) cells, which are central to the pathogenesis of pulmonary fibrosis (搜索). The peptide therapeutic is designed for subcutaneous administration and works as an agonist of β1 integrin (搜索), a transmembrane protein critical for AEC2 function.
"ORJ-001 (搜索) was designed with this biology in mind and may provide, for the first time, a therapeutic intervention that repairs and reverses fibrosis and promotes disease modification," said Sujay Kango, Chief Executive Officer of Oorja Bio (搜索).
The β1 integrin (搜索) pathway is essential for regulating AEC2 cell fate, including senescence and differentiation into mature alveolar epithelial type 1 cells, which line the majority of the alveolar surface. The therapy also influences inflammatory signaling pathways implicated in fibrosis, potentially offering both disease-halting and repair-promoting effects.
Promising Preclinical Results
In extensive preclinical studies using validated animal models of pulmonary fibrosis (搜索), ORJ-001 (搜索) demonstrated the ability to reverse established fibrosis and regenerate normal lung and alveolar morphology. Additional preclinical studies showed positive effects on biomarkers of tissue remodeling associated with fibrotic disease progression.
These results address a critical gap in IPF (搜索) treatment, where currently approved therapies can slow disease progression but do not prevent continued decline in lung function or reverse existing damage.
Experienced Leadership Team
Oorja Bio (搜索) is led by a founding team with proven track records in pulmonary drug development. CEO Sujay Kango and Chief Medical Officer Janethe Pena, MD, PhD, previously collaborated at Acceleron Pharma (搜索) to lead the product strategy and clinical development of sotatercept, a first-in-class biologic for pulmonary arterial hypertension (搜索) that became the first drug approved for PAH (搜索) treatment in ten years.
Kango was most recently CEO of Tmunity Therapeutics (搜索), which was acquired by Gilead, and previously served as Chief Commercial Officer at Acceleron, where he was responsible for the commercialization of blockbuster therapy Reblozyl®. Dr. Pena continued her work with sotatercept at Merck following Acceleron's acquisition and previously held leadership roles at Bayer (搜索), working on pivotal trials for Adempas® approval.
Addressing Significant Unmet Need
IPF (搜索) affects more than 150,000 adults in the United States and is characterized by progressive fibrosis of lung tissue, leading to declining lung function, respiratory failure, and ultimately death. The disease carries a poor prognosis, with an average life expectancy of three to five years after diagnosis.

The Series A funding will support ORJ-001 (搜索)'s clinical advancement and enable Oorja Bio (搜索) to build a pipeline of assets addressing other diseases involving fibrosis or cardiopulmonary function. The company acquired global rights to ORJ-001 as part of its in-licensing strategy for therapeutic innovation in fibrosis, with plans to target the underlying pathophysiology of fibrotic diseases beyond symptom management.
