Optigo Biotherapeutics Appoints Industry Veteran Andreas Wallnöfer to Board as Long-Acting Anti-VEGF Advances to IND-Enabling Studies
核心洞察
Optigo Biotherapeutics (搜索) has appointed Dr. Andreas Wallnöfer, a pharmaceutical R&D veteran with over 25 years of experience, to its Board of Directors as the company advances its lead anti-VEGF (搜索) program toward IND-enabling studies.
The company's lead candidate XPK-640 (搜索) utilizes a proprietary hyaluronic acid-anchoring platform that demonstrated potential to extend aflibercept durability by at least 10-fold in cynomolgus monkey studies, potentially enabling once-yearly dosing.
Wallnöfer previously served as a partner at Jeito Capital (搜索) where he led early investment in EyeBio (搜索), which was acquired by Merck (搜索) for $3 billion in 2024, and held senior R&D roles at Roche including contributing to Vabysmo development.
Optigo Biotherapeutics (搜索), a Vancouver-based ophthalmology company developing next-generation retinal disease treatments, has appointed Dr. Andreas Wallnöfer to its Board of Directors as the company advances its lead anti-VEGF (搜索) program toward Investigational New Drug (IND)-enabling studies. The strategic appointment comes as Optigo prepares to enter clinical development with its long-acting anti-VEGF candidate XPK-640 (搜索) for neovascular age-related macular degeneration (搜索).
Industry Veteran Brings Extensive R&D and Investment Experience
Dr. Wallnöfer brings more than 25 years of leadership in pharmaceutical R&D and biotech investment to Optigo's board. Most recently serving as a Partner at Jeito Capital (搜索), he was one of the initial lead investors in EyeBio (搜索), which Merck (搜索) subsequently acquired in 2024 for a total deal value of $3 billion. Prior to Jeito, he served as a General Partner at BioMedPartners (搜索), where he was involved in multiple M&A acquisitions.
Earlier in his career, Wallnöfer held senior positions on the R&D Executive Leadership Team at F. Hoffmann-La Roche AG, serving as Global Head of Clinical Research & Exploratory Development and Head of Cardiovascular & Metabolism R&D. During his tenure at Roche, he played a key role in the Roche/Genentech integration and contributed to the development of Vabysmo (faricimab-svoa (搜索)), one of the most successful ophthalmology launches in recent years.
"Andreas is exactly the strategic leader and industry veteran we were looking for," said Dr. Henning Afflerbach, Chair of Optigo's Board. "His singular blend of operational experience — from bench to boardroom — and his track record of guiding programs through to successful exits will be invaluable as we move into clinical development."
Breakthrough Platform Technology Shows Promise
Wallnöfer expressed enthusiasm about Optigo's proprietary hyaluronic acid (HA)-anchoring platform, calling it "one of the most promising innovations in retinal drug design" he has seen. The platform's unprecedented half-life extension has the potential to redefine treatment durability and transform the standard of care for retinal diseases (搜索).
The company's lead candidate XPK-640 (搜索), a long-acting anti-VEGF (搜索) treatment, is moving toward pre-IND work and Good Laboratory Practice safety testing for neovascular age-related macular degeneration (搜索). Recently completed cynomolgus monkey pharmacokinetic and tolerability studies demonstrated that XPK-640 has the potential to extend the durability of aflibercept by at least 10-fold.
According to Dr. Houman David Hemmati, co-founder and chief medical officer of Optigo, this could translate to once-yearly dosing intervals in patients, or perhaps even less frequently, likely without the burden and compliance risks of monthly loading doses.
Addressing Critical Unmet Medical Need
The appointment comes at a crucial time as Optigo prepares to address a significant unmet need in retinal disease treatment. Real-world data show that nearly 40% of eyes discontinue anti-VEGF (搜索) treatment due to injection burden after the first year, highlighting the critical need for longer-acting therapies.
"The greatest unmet need for patients with chronic retinal neovascular diseases has been reducing the intravitreal injection burden," Hemmati explained. "XPK-640 (搜索) has the potential to be the first drug that addresses this need without the safety risks or efficacy concerns of devices, gene therapies, injectable polymers, or small molecule kinase inhibitors."
CEO Ali Ardakani emphasized the personal motivation behind the company's mission, sharing that his grandmother's experience with wet AMD (搜索) and monthly injections inspired the development of longer-acting treatments. "For patients who live in fear of losing their vision, every new advance matters," Ardakani said. "Reaching the IND-enabling stage brings us one step closer to a long-acting therapy that could ease that burden and change what's possible for millions of people."
Clinical Timeline and Development Strategy
The company expects to enter clinical trials within 12 to 15 months with this new class of anti-VEGF (搜索) product. Optigo's preclinical data suggest the platform can maintain therapeutic levels in the eye for far longer than existing treatments, potentially up to 12 months.
"Andreas joining our Board signals the next phase of growth for Optigo — we are accelerating our development timeline to advance our lead program toward IND-enabling toxicology in preparation for first-in-human trials," said Ardakani. "Adding Andreas to the team underscores our commitment to strong governance and disciplined execution."
Leveraging its proprietary long-acting HA-binding anchoring platform, Optigo has engineered a pipeline of assets designed to address unmet needs across multiple retinal indications, positioning the company to potentially transform treatment paradigms in ophthalmology.
