PAQ Therapeutics Initiates First-in-Human Trial of Pan-KRAS Degrader PT0511, Secures $77 Million in Series B Funding
核心洞察
PAQ Therapeutics (搜索) has dosed the first patient in a Phase 1 clinical trial evaluating PT0511 (搜索), a pan-KRAS (搜索) degrader designed to target multiple oncogenic KRAS variants in advanced solid tumors (搜索).
The company completed a Series B extension bringing total Series B financing to $77 million to support clinical advancement of PT0511 (搜索) and ongoing development of PT0253, their KRAS G12D (搜索) degrader.
PT0511 (搜索) represents an expansion beyond single-mutation KRAS (搜索) targeting and aims to address key limitations observed with existing KRAS or pan-RAS inhibitor therapies through targeted protein degradation.
PAQ Therapeutics (搜索), a clinical-stage oncology company focused on targeted protein degradation therapies for KRAS-driven cancers (搜索), has achieved two significant milestones: dosing the first patient in a Phase 1 clinical trial of PT0511 (搜索), their pan-KRAS (搜索) degrader, and completing a Series B extension that brings total Series B financing to $77 million.
Clinical Trial Initiation
PT0511 (搜索) is designed as a pan-KRAS (搜索) degrader that targets multiple oncogenic KRAS variants, representing a broader approach compared to therapies targeting single KRAS mutations. The Phase 1 study is a first-in-human, open-label, dose-escalation trial that will evaluate the safety, tolerability, pharmacokinetics, and preliminary anti-tumor activity of PT0511 in patients with advanced solid tumors (搜索) harboring KRAS alterations.
"The initiation of clinical dosing with PT0511 (搜索) expands our clinical portfolio beyond single-mutation KRAS (搜索) targeting," said Andrew Krivoshik, MD, PhD, Chief Medical Officer of PAQ Therapeutics (搜索). "A pan-KRAS degradation approach has the potential to address key limitations for patients observed with existing KRAS- or pan-RAS inhibitor therapies."
Financing and Pipeline Development
The Series B extension included participation from both existing and new investors, strengthening PAQ's financial position to advance multiple clinical programs. The proceeds will support the ongoing Phase 1 development of PT0253, PAQ's KRAS G12D (搜索) degrader, alongside the clinical advancement of PT0511 (搜索).
"Completing this Series B extension and dosing the first patient in our PT0511 (搜索) Phase 1 study represent important milestones for PAQ," said Nan Ji, PhD, Chief Executive Officer of PAQ Therapeutics (搜索). "Together, these achievements highlight our continued execution against a multi-program clinical strategy and our focus on addressing significant unmet need across KRAS-driven cancers (搜索)."
Therapeutic Approach
PAQ's strategy centers on leveraging targeted protein degradation to achieve what the company describes as deep and selective suppression of oncogenic signaling while maintaining favorable safety and combinability profiles. This approach differentiates their KRAS (搜索) pipeline from existing therapeutic options in the oncology landscape.
The company positions itself as developing best- and first-in-class KRAS (搜索) degraders specifically for lethal cancers that currently lack effective treatment options, addressing a significant gap in cancer therapeutics.
