Parabolus Medicines (搜索) is advancing a novel class of alpha-helical peptide drugs against a target the pharmaceutical industry has spent three decades failing to drug. The company's lead candidate, zolocatetide (搜索), binds the beta-catenin (搜索)/TCF (搜索) transcription factor complex — a flat, pocket-free intracellular protein surface that conventional small molecules and biologics cannot grip — and has produced a 74% objective response rate in desmoid tumors (搜索), a rare soft-tissue cancer with no approved therapy beyond gamma secretase inhibitors.
The company, led by CEO Mathai Mammen, went public earlier in 2026, raising $770 million in what is described as the largest venture-backed biotech IPO on record. Mammen laid out the company's strategy and clinical data in a September 2026 interview on The Long Run.
The undruggable target problem
The central scientific premise is that the pharmaceutical industry has only ever been able to drug a minority of disease-relevant proteins. Small molecules work by occupying well-defined pockets or clefts on protein surfaces, but Mammen estimates that 80% or more of intracellular proteins lack such pockets. Transcription factors, E3 ligases, and many signaling proteins present flat, smooth surfaces that conventional chemistry cannot grip.
Beta-catenin (搜索) is a transcription factor that, when dysregulated, drives cell proliferation and is implicated in an estimated 10% of all cancers. The specific complex Parabolus targets — beta-catenin bound to TCF (搜索) — has been a sought-after drug target since 1990. Mammen said he personally attempted it at both Merck (搜索) and Johnson & Johnson, as did essentially every major pharmaceutical company, and all failed. "The protein is perfectly smooth," Mammen said, with no cavities for small molecules to occupy.
The alpha-helical peptide platform
Parabolus's approach builds on an observation from nature: the membrane-spanning domains of essentially all proteins are alpha helices. These coiled structures can cross cell membranes because their internal hydrogen bonds are "self-satisfied" — they do not need to shed water to pass through a lipid bilayer. Founder Greg Verdine, who started the company (originally as Fog Pharma) out of Harvard roughly a decade ago, reasoned that a stabilized alpha helix could itself act as a drug.
The company's two technical inventions address the limitations of natural amino acids. First, it stabilizes peptides in alpha-helical conformation through chemical linkages that bridge successive turns of the helix. Second, because the helix is already stabilized, the company can freely substitute unnatural amino acids throughout the sequence — over 3,000 synthetic building blocks by Mammen's count, creating what he describes as astronomical sequence diversity (3,000 to the 20th power possible combinations for a 20-residue peptide).
The approach contrasts with an earlier Verdine-founded company, Aileron, which used a single covalent "staple" to lock the helix. That effect was local — moving two or three amino acids away from the staple caused the peptide to revert to non-helical behavior — which limited the number of unnatural amino acids that could be used. Parabolus's hundreds of stabilization chemistries, Mammen argues, are what transformed a proof of concept into a drug platform.
Clinical data in desmoid tumors
The first clinical foray was in colorectal cancer (搜索), where APC mutations are the truncal, initiating mutation. The trial achieved what Mammen described as the critical de-risking step: the drug was safe and tolerable at doses that hit the target hard, with no dose-limiting toxicity, and the company observed disease stabilization, ctDNA knockdown, and tumor cavitation. However, in fifth- and sixth-line colorectal cancer patients whose tumors had accumulated dozens of additional mutations, efficacy was not deep enough to justify continued development.
"We could pursue this, it could be in late-line CRC, a monotherapy approval, but I didn't feel that we were going to add meaningfully to the armamentarium used to treat colorectal cancer (搜索)," Mammen said. "We would do something, but it wouldn't be significant. It wouldn't be worth the time and energy to either us or the patients."
The company pivoted to "purer" cancers driven more singularly by the beta-catenin (搜索) pathway. Desmoid tumors (搜索) are rare, locally invasive soft-tissue tumors that can cause decades of intractable pain and dysfunction; surgery is generally not curative because clean margins are nearly impossible to achieve.
The zolocatetide (搜索) data, presented around the time of the IPO, showed a 74% objective response rate across 38 patients who had typically progressed on prior therapy. Mammen emphasized that responses were durable — patients have been on therapy for up to three years — and that essentially all tumors were shrinking, suggesting the 74% figure understates the uniformity of response. The drug is currently administered intravenously, with a subcutaneous, self-administered formulation using a device similar to the semaglutide pen planned for clinical work in 2027.
Safety concerns that had worried the field did not materialize. Novartis's earlier work on porcupine, a protein upstream in the Wnt pathway, had shown profound bone effects and taste disturbance (dysgeusia). Parabolus saw neither at clinically relevant doses; bone effects were "extraordinarily mild" and manageable with standard osteoporosis drugs, and dysgeusia appears to have been an off-target effect of the porcupine approach rather than an on-target Wnt/beta-catenin (搜索) blockade effect.
Expansion strategy and platform economics
The expansion map is a sequential march through indications where beta-catenin (搜索) is the dominant driver. A Phase 3 start in desmoid tumors (搜索) is planned for the first half of 2027. Early data in familial adenomatous polyposis (搜索) (FAP), adamantinomatous craniopharyngioma (搜索) (ACP), and hepatocellular carcinoma (搜索) (HCC) were disclosed at the IPO — the first two indications have no approved medicine, and existing HCC therapies are described as poor. He did not announce a formal program but described it as "very logical for us to go down that road."
The IPO proceeds fund three priorities: expanding zolocatetide (搜索) across the indication list, advancing two undisclosed prostate cancer programs, and building out the platform beyond oncology through a collaboration with Regeneron. Mammen's stated ambition is to build "the next Regeneron, Vertex, or Amgen."
On the data-science front, Parabolus applies generative models with reinforcement learning trained on its proprietary helicon peptide dataset spanning over 3,000 unnatural building blocks — a dataset Mammen argues no AI-native drug discovery company could replicate, since their models are trained on natural amino acid chemistry. The company also collects pre- and post-dosing biopsies, maintains hundreds of patient-derived xenograft models, and applies computation to identify genetic and imaging signatures that predict response, with a long-term goal of tumor-agnostic approval based on a molecular signature rather than anatomical site of origin.
What to watch
The next 12–24 months will determine whether Parabolus is a platform story or a one-drug story. Key developments include the Phase 3 desmoid tumor start in H1 2027, the subcutaneous formulation clinical work planned for 2027, the two undisclosed prostate cancer programs, the Regeneron collaboration's first disclosed targets outside oncology, and any formal announcement of a RAS inhibitor combination study in colorectal cancer (搜索). The tumor-agnostic basket study, if successful, would be the clearest validation that the platform can generalize across indications.
