University of Pennsylvania spinout Linnaeus Therapeutics (搜索) has secured $22 million in federal funding from the Advanced Research Projects Agency for Health (ARPA-H (搜索)) to advance clinical testing of its novel anti-aging (搜索) drug candidate. The Haddonfield-based biotech is among seven research teams nationwide selected for ARPA-H's PROactive Solutions for Prolonging Resilience (PROSPR) program, which aims to prevent age-related diseases and extend healthspan.
GPER Targeting Offers Estrogen Benefits Without Hormone Risks
Linnaeus' lead compound, LNS8801, represents a breakthrough approach to harnessing estrogen's protective effects without traditional hormone therapy risks. The once-daily pill works by selectively binding to G protein-coupled estrogen receptor (搜索) (GPER (搜索)), allowing the drug to activate beneficial estrogen pathways while avoiding reproductive system effects.
"The drug's design can target this particular estrogen receptor without activating other processes that estrogen plays a hand in," explained Christopher Natale, co-founder and chief scientific officer. The selective targeting means patients avoid traditional estrogen-related effects such as feminizing changes like breast development, since LNS8801 doesn't bind to estrogen receptor alpha (搜索) or beta.
From Melanoma Research to Aging Applications
The company's origins trace back to Natale's doctoral research at Penn, where he investigated why women with melanoma (搜索) consistently showed better outcomes than men. Research had demonstrated that women with skin cancer had lower mortality and recurrence rates, with men diagnosed between ages 15-39 being 55% more likely to die of melanoma than women in the same age group.
Natale's laboratory experiments revealed that estrogen inhibited melanoma (搜索) cell growth through GPER (搜索) activation. "GPER was a fairly novel estrogen receptor at the time," Natale noted. This discovery led to the 2016 founding of Linnaeus Therapeutics (搜索) alongside Penn dermatologist Todd Ridky.

LNS8801 has been tested in melanoma (搜索) clinical trials since 2019, receiving both Orphan Drug and Fast Track designations from the FDA. Early trials involving approximately 100 cancer patients reported no serious adverse events, establishing a favorable safety profile.
Beyond cancer treatment, trial participants experienced unexpected health improvements. Some patients saw improvements in cholesterol levels, while those with elevated hemoglobin A1C levels related to diabetes (搜索) and high blood pressure also showed positive responses. These observations prompted CEO Patrick Mooney to recognize the drug was "potentially much bigger than cancer and could benefit people with diseases of aging (搜索), writ large."
Expanding Research Into Aging and Healthspan
The five-year ARPA-H (搜索) funding will enable Linnaeus to launch clinical trials specifically testing LNS8801's ability to preserve physical and mental abilities in older adults. This research addresses a critical need, as more than 90% of adults over 65 have a chronic condition, and nearly 80% have two or more, according to the Centers for Disease Control and Prevention.
Sarah Lindsey, a pharmacology professor at Tulane University who tested the drug in vascular health experiments, found that LNS8801 reduced arterial stiffness in mice experiencing the equivalent of menopause. "We know that estrogen has a lot of anti-aging (搜索) properties in general," Lindsey observed, noting the drug's aging applications were "not that far-fetched."
Strong Financial Position Supports Expansion
Linnaeus has demonstrated robust fundraising capabilities across multiple channels. The company recently closed a $4.5 million Series B extension to support its next clinical trial and team expansion. "We're using this money that we just raised to start the clinical trial, but we'll use the ARPA-H (搜索) money to follow the science and see if we can prolong resilience," Mooney explained.
Scientific Validation and Future Outlook
The ARPA-H (搜索) selection validates Linnaeus' approach of using "observed sex differences to identify mechanisms like GPER (搜索) activation that link tumor suppression, vascular health, metabolism, and brain function." However, experts urge careful development approaches.
Deborah Clegg, vice president for research at Texas Tech University Health Sciences Center El Paso, cautioned that GPER (搜索) activation "is not just a light switch" but requires nuanced understanding. She emphasized the importance of activating the right cell types at appropriate levels to maximize benefits, with dosing potentially dependent on individual factors and aging (搜索) spectrum position.
The company's research builds on evolving understanding of estrogen therapy, as recent studies suggest estrogen could reduce risks of breast cancer, cardiovascular disease (搜索), bone fractures, and cognitive decline. The FDA recently removed black box warnings on hormone therapies for menopausal women after reviews found them safe and effective.
