Primmune Therapeutics (搜索) announced the completion of an additional $8.6 million close of its Series B financing round, bringing the total Series B raise to $23.3 million. The financing included participation from Bioqube Ventures (搜索), Oberland Capital (搜索), and Samsara Biocapital (搜索), and will support continued clinical development of the company's lead immunotherapy candidate PRTX007.
The biotech company plans to use the funds to initiate Study PRTX007-003, a Phase 2 neoadjuvant efficacy study evaluating PRTX007 in combination with standard-of-care anti-PD-1 therapy (搜索) in patients with Stage III resectable melanoma (搜索). The study will be conducted entirely in Australia by Primmune Therapeutics (搜索) Pty Ltd. in conjunction with Novotech (搜索) acting as the in-country clinical research organization.
Novel TLR7 Agonist Design
PRTX007 represents a novel approach to toll-like receptor 7 (TLR7 (搜索)) agonism, designed in-house at Primmune to address limitations that have hindered previous TLR agonist development. The orally administered, systemically acting small molecule is engineered to functionally tune immune signaling toward an IRF7 (搜索)-driven poly-interferon response while avoiding the NF-KB (搜索)-mediated pro-inflammatory cytokine signaling that has limited the utility of other systemically acting TLR7, TLR7/8, and TLR8 agonists.

Clinical Development Progress
PRTX007 has demonstrated encouraging safety and pharmacodynamic profiles in early-stage clinical testing. The compound has been administered to over 100 healthy human volunteers across two separate Phase 1 clinical studies (Study PRTX007-001 and Study PRTX007-002). In these studies, PRTX007 successfully drove the desired systemic IRF7 (搜索) poly-interferon response without triggering the undesired NF-KB (搜索) pro-inflammatory response.
The Phase 1 studies showed that PRTX007 was generally well-tolerated with no serious adverse events (SAEs) reported across the study population. This safety profile supports the company's advancement into the planned Phase 2 efficacy study in melanoma (搜索) patients.
