Protagonist Therapeutics Advances Multiple Drug Programs with Strong Financial Position Through 2028

Protagonist Therapeutics reported third quarter 2025 financial results showing a strong cash position of $678.8 million, representing an increase from $559.2 million at the end of 2024. The company expects this funding to support operations through at least the end of 2028 as it advances multiple drug candidates through clinical development.
Regulatory Milestones for Lead Programs
The company achieved significant regulatory progress with icotrokinra, a first-in-class oral IL-23 receptor (搜索) antagonist developed in partnership with Johnson & Johnson. Following the New Drug Application (NDA) submission to the U.S. FDA in July 2025 for treating adults and adolescents with moderate to severe plaque psoriasis (搜索), Protagonist and Johnson & Johnson submitted an application to the European Medicines Agency (EMA) in September 2025.

Recent clinical data presentations have demonstrated icotrokinra's efficacy across multiple conditions. At the 2025 ACG Annual Scientific Meeting, Week 28 results from the Phase 2b ANTHEM-UC study showed 31.7% of patients with moderately to severely active ulcerative colitis (搜索) achieved clinical remission, with 38.1% showing endoscopic improvement versus placebo.
Breakthrough Designation for Rusfertide
Rusfertide, a subcutaneous injectable hepcidin mimetic co-developed with Takeda Pharmaceuticals (搜索), received breakthrough therapy designation from the FDA for treating erythrocytosis in patients with polycythemia vera (搜索) (PV). The designation was granted on August 25, 2025, and Takeda is expected to file the U.S. NDA for rusfertide in the fourth quarter of 2025.
The drug will be featured in four presentations at the 67th Annual American Society of Hematology meeting in December 2025, including an oral presentation of durability of response and safety results through week 52 from the Phase 3 VERIFY study.
Pipeline Expansion and Early-Stage Programs
Protagonist initiated dosing of the first patient in the Phase 1 trial of PN-881, a first-in-class oral IL-17 (搜索) peptide antagonist. The study (NCT07153146) is evaluating safety, tolerability, pharmacokinetics, and pharmacodynamics in healthy adult participants.
The company is also advancing triple-GLP (搜索)/GIP (搜索)/GCG (搜索) agonists PN-477sc (搜索) and PN-477o (搜索) through IND-enabling studies. Clinical study initiation of PN-477sc is anticipated by mid-2026, with PN-477o expected to begin trials in the second half of 2026. Additionally, Protagonist plans to nominate a development candidate from its oral hepcidin program by year end.
Financial Performance
For the third quarter of 2025, Protagonist reported a net loss of $39.3 million, or $0.62 per share, compared to a net loss of $33.2 million, or $0.54 per share, in the same period of 2024. License and collaboration revenue remained stable at $4.7 million for both quarters.
Research and development expenses increased by $4.0 million to $40.0 million for the third quarter, primarily due to increases in drug discovery and pre-clinical research expenses. General and administrative expenses rose by $1.0 million to $11.1 million, mainly from increased professional services costs.
