Health Canada has approved Sephience for the treatment of hyperphenylalaninemia (搜索) (HPA) in adult and pediatric patients one month old and older with sepiapterin-responsive phenylketonuria (搜索). The drug is now commercially available in Canada, providing a new treatment option for patients who must maintain strict phenylalanine-restricted diets.

The Health Canada approval is based on evidence of significant efficacy and safety from the phase 3 APHENITY trial as well as durability of treatment effect in the APHENITY long-term extension study.
Japan Approval Marks Company Milestone
In a separate regulatory achievement, the Japanese Ministry of Health, Labor and Welfare (搜索) (MHLW) approved Sephience for PKU treatment in children and adults, with the label including individuals of all ages and the full spectrum of disease severity. This represents PTC Therapeutics' first product approval in Japan.

PTC will now engage in pricing discussions in Japan, which are expected to conclude in Q1 2026, with launch occurring shortly thereafter.

Phenylketonuria (搜索) is a rare, inherited metabolic disease characterized by the body's inability to break down phenylalanine, an essential amino acid. If left untreated or poorly managed, phenylalanine can build up to harmful levels, causing severe and irreversible disabilities including permanent intellectual disability, seizures, delayed development, memory loss, and behavioral and emotional problems.
Newborns with PKU initially show no symptoms, but the condition is progressive, and damage from toxic phenylalanine levels in early life is irreversible. Diagnosis typically occurs through newborn screening programs, emphasizing the importance of early intervention.

With these latest approvals, Sephience is now approved in the European Economic Area, the United States, Canada, and Japan, representing a comprehensive global regulatory achievement for PTC Therapeutics.
