Quanta Therapeutics Advances KRAS G12D Inhibitor QTX3034 to Dose Expansion Phase Following Positive Safety Data
核心洞察
Quanta Therapeutics (搜索) has initiated enrollment in dose expansion cohorts for its Phase 1 clinical trial of QTX3034, an oral G12D-preferring multi-KRAS (搜索) inhibitor, following favorable safety and preliminary efficacy data.
The dose expansion phase will evaluate QTX3034 in patients with KRASG12D (搜索)-mutant pancreatic, colorectal, and endometrial cancers as both monotherapy and in combination with cetuximab.
The company appointed 25-year industry veteran Vanessa L. Jacoby as Chief Business Officer and Chief Financial Officer to support its clinical development and growth strategy.
Quanta Therapeutics (搜索) has initiated enrollment in dose expansion cohorts for its Phase 1 clinical trial of QTX3034, an oral G12D-preferring multi-KRAS (搜索) inhibitor, marking a significant milestone in the development of treatments for KRASG12D (搜索)-mutant cancers. The advancement to dose expansion is supported by favorable safety and encouraging preliminary efficacy data from the dose escalation portion of the trial.
The privately-held clinical-stage biopharmaceutical company simultaneously announced the appointment of Vanessa L. Jacoby as Chief Business Officer and Chief Financial Officer, bringing more than 25 years of financial and strategic leadership experience in the biopharmaceutical industry to support the company's next phase of development.
Clinical Trial Progress and Design
The Phase 1 clinical trial is evaluating QTX3034 both as monotherapy and in combination with cetuximab in patients with solid tumors (搜索) harboring KRASG12D (搜索) mutations. The dose expansion cohorts are specifically enrolling patients with KRASG12D-mutant pancreatic, colorectal, and endometrial cancers.
"The opening of dose expansion cohorts in the Phase 1 clinical trial of QTX3034 is an important milestone for the program and company, signifying early positive safety and anti-tumor activity as a monotherapy and in combination," said Perry Nisen, MD, PhD, Chief Executive Officer of Quanta. "We look forward to progressing the program and presenting data at a major medical meeting later this year."
The Phase 1 clinical endpoints include safety and tolerability, determination of the maximum tolerated dose/recommended Phase 2 dose, pharmacokinetic properties, antitumor activity, and molecular markers. The clinical trial is being conducted at clinical sites in the US under the identifier NCT06227377.
Targeting Previously Undruggable KRAS Mutations
QTX3034 is a multi-KRAS (搜索) inhibitor with G12D-preferring activity designed to address one of the most challenging targets in oncology. KRAS mutations are among the most prevalent drivers of cancer, with the G12D mutation representing a significant subset of these oncogenic alterations.
"I am delighted to be joining Quanta at this pivotal stage of development as the company continues to advance novel small molecule cancer medicines targeting previously undruggable KRAS (搜索) mutations with the potential to address the most prevalent drivers of cancer," said Ms. Jacoby. "I look forward to partnering with the team to support the advancement of our pipeline and build a strong financial foundation for future growth."
Leadership Appointment and Strategic Direction
Ms. Jacoby brings extensive experience from her previous roles in the biopharmaceutical industry. Prior to joining Quanta, she served as Chief Financial Officer of Shoreline Biosciences (搜索), where she played a key role in the marketing and closing of Shoreline's $140M Series B round, as well as the establishment of partnerships with Kite Pharma (搜索) and BeiGene (搜索), both of which included meaningful upfront payments and a combined total deal value of $3.7 billion.
Previously, Ms. Jacoby served as Chief Accounting Officer of Avidity Biosciences (搜索), where she led corporate finance and planning activities and played a key role in the company's initial public offering and crossover financing. She also held positions of increasing responsibilities at PharmAkea, which was acquired by Galecto in 2019, Brain Cells, Artes Medical and Verenium.
Company Platform and Pipeline
Quanta Therapeutics (搜索) focuses on developing novel small molecule cancer medicines by selectively targeting protein-protein interactions that are key to oncogenic RAS (搜索) activity. The company's unique high-throughput platform applies Second Harmonic Generation (SHG) optical technology to identify allosteric modulators of protein complexes.
The company's KRAS (搜索) inhibitor pipeline includes QTX3034, currently in Phase 1 clinical trial as monotherapy and in combination with cetuximab, and QTX3544, a multi-KRAS inhibitor with G12V-preferring activity, currently in a Phase 1 clinical trial as monotherapy and in combination in patients with KRASG12V (搜索)-driven solid tumors (搜索).
By applying innovative medicinal chemistry and its unique protein conformation detection technology, Quanta aims to advance differentiated, next-generation RAS (搜索) programs that address the resistance paradigms of targeted therapy in oncology.
