Replicate Bioscience (搜索), a clinical-stage company pioneering self-replicating RNA (srRNA) technology, has entered into a collaboration agreement with Instituto Butantan, Brazil's leading non-profit producer of immunobiologic products, to develop and commercialize the novel srRNA rabies vaccine RBI-4000 in Latin America. The partnership grants Instituto Butantan exclusive rights to the vaccine in Brazil and other Latin American countries.
Clinical Performance Demonstrates Promise
In Phase 1 clinical studies conducted by Replicate, RBI-4000 demonstrated a high therapeutic index showing durable bioactivity at low doses accompanied by mild to moderate reactogenicity, contrasting favorably with other RNA-based vaccine technologies. The vaccine has shown protective levels of immunity at doses lower than any other reported mRNA or srRNA vaccine in Phase I trials.
Partnership Structure and Financial Terms
Under the agreement terms, Instituto Butantan will fund the development of RBI-4000 for both post-exposure and pre-exposure prophylaxis applications. Replicate will provide GMP material for clinical trials and transfer the manufacturing process to Butantan's mRNA vaccine production facility. The company remains eligible to receive tiered royalties on future product sales in Latin America while retaining rights for RBI-4000 in markets outside the region and access to all data generated under the agreement.
Addressing Critical Medical Need
Rabies represents a significant global health challenge, endemic in over 150 countries with a 99% mortality rate if left untreated, causing 59,000 deaths annually. Deaths due to rabies are fully preventable if exposed individuals receive an effective vaccine before the virus spreads to the central nervous system and clinical symptoms appear. Current rabies vaccines require 3 to 5 doses and face supply shortages due to complicated manufacturing processes, limiting accessibility and increasing administration complexity in resource-limited settings.
The World Health Organization has included rabies in its 2021-2030 Roadmap for the global control of neglected tropical disease. Additionally, as part of the 21st Century Cures Act, the U.S. Food and Drug Administration included rabies in its Tropical Disease Priority Review Voucher Program List.
Technology Advantages
The collaboration will accelerate improvement of cold chain storage and thermostability, which may bolster adoption of srRNA technology and support a cost advantage versus conventional vaccine technologies. Replicate's srRNA platform aims to overcome shortcomings of existing mRNA approaches with potential improvements in bioactivity, induction of more potent immune responses, and improved therapeutic index.
