Rezolute's Ersodetug Fails Phase 3 Trial for Congenital Hyperinsulinism, Stock Plunges 88%
核心洞察
Rezolute's Phase 3 sunRIZE trial evaluating ersodetug for congenital hyperinsulinism (搜索) failed to meet its primary endpoint, showing no statistically significant reduction in hypoglycemia (搜索) events compared to placebo.
Despite achieving a 45% reduction in hypoglycemia (搜索) events at the highest dose, this was not significantly different from the 40% improvement observed in the placebo group.
The company's stock crashed 88.76% to $1.23 following the disappointing results, with trading volume surging to 82.25 million shares compared to the typical 1.597 million.
Rezolute Inc. experienced a devastating 88.76% stock decline to $1.23 on Thursday following the release of disappointing Phase 3 trial results for its lead drug candidate ersodetug in treating congenital hyperinsulinism (搜索). Trading volume exploded to 82.25 million shares, more than 50 times the average daily volume of 1.597 million shares.
Trial Results Fall Short of Expectations
The Phase 3 sunRIZE study failed to meet its primary endpoint, which assessed the change in average weekly hypoglycemia (搜索) events by self-monitored blood glucose. While patients receiving the highest dose of ersodetug (10 mg/kg) experienced an approximate 45% reduction in hypoglycemia events, this improvement was not statistically significant when compared to the placebo group, which showed a 40% improvement.
The study also missed its key secondary endpoint measuring the change in average daily percent time in hypoglycemia (搜索) by continuous glucose monitoring (CGM). At the 10 mg/kg dose, ersodetug demonstrated an approximate 25% reduction in time spent in hypoglycemia, but this was not statistically significant compared to placebo, which increased by approximately 5%.
Drug Achieved Target Concentrations Across Age Groups
Despite the efficacy shortfall, ersodetug achieved target drug concentrations across all age groups studied when administered at doses of 5 and 10 mg/kg every other week for three doses, followed by every 4 weeks over the remainder of the 24-week treatment duration.
Safety Profile Generally Favorable with Notable Exceptions
Safety observations from the study were generally favorable and, according to the company, support safe use of ersodetug in both pediatric and adult patients. However, two study participants experienced serious hypersensitivity reactions that led to early discontinuation of the study drug.
Understanding Congenital Hyperinsulinism
Congenital hyperinsulinism (搜索) is a rare genetic disorder that causes the pancreas to secrete excessive amounts of insulin, leading to dangerously low blood sugar levels (hypoglycemia (搜索)) in infants and children. This condition represents a significant unmet medical need in pediatric endocrinology.
Pipeline Continues with Tumor Hyperinsulinism Study
While the congenital hyperinsulinism (搜索) program has encountered this major setback, Rezolute continues to advance its Phase 3 upLIFT study evaluating ersodetug for tumor-associated hyperinsulinism (搜索). The company expects topline results from this ongoing trial in the second half of 2026.
