Genentech, Roche's subsidiary, announced Tuesday that it is expanding its Holly Springs manufacturing plant investment from the initial $700 million commitment made in May. The 700,000-square-foot facility will now create over 500 jobs, an increase of 100 positions from the original plan of 420 jobs.
Strategic Challenge to GLP-1 Market Leaders
"Our intention is to be a challenger to those two big players," said Paul Bezy, Roche's global head of high-volume supply chain operations, referring to the current obesity (搜索) drug duopoly. "And we've got the confidence in the pipeline that we're building to be that challenger."
The facility expansion directly supports Roche's ambitions to become a top three player in the obesity (搜索) market, where Eli Lilly and Novo Nordisk currently hold the leading positions with their GLP-1 (搜索) treatments. Novo Nordisk produces Ozempic and Wegovy, while Eli Lilly manufactures Zepbound and Mounjaro.

Roche expects to present Phase III-enabling data on five obesity (搜索) candidates this year, including results from a midphase study of CT-388. The injectable CT-388 is a dual GLP-1 (搜索)/GIP (搜索) receptor agonist with the same mechanism of action as Lilly's market-leading Mounjaro and Zepbound.
At an investor event in September, Roche CEO Teresa Graham explained that the company will initially use internal and external capacity to support clinical trials of CT-388 and the drug's initial launch, eventually transitioning to a primarily internal supply chain.
Graham emphasized that the purpose of building Holly Springs is to ensure Roche has a "large volume throughput device facility." She noted that devices have been identified as the biggest bottleneck for current obesity (搜索) players, stating that analysts "can be very confident that we will be entering with devices in the places where devices are necessary."

The massive market potential has attracted significant investment from multiple pharmaceutical companies beyond the current leaders, with Pfizer and others also investing heavily in weight loss drug development.
Facility Timeline and Economic Impact
The Holly Springs plant, Genentech's first East Coast production facility, is expected to become operational in 2029. The company broke ground on the site in August after receiving substantial incentive packages from local and state governments.
North Carolina awarded Roche incentives worth up to $13 million contingent on Genentech meeting its hiring and investment targets, while Holly Springs and Wake County contributed a $33.5 million performance-based package.
Regional Pharmaceutical Hub Development
Holly Springs has emerged as a significant pharmaceutical manufacturing hub, hosting facilities from CSL Seqirus, Fujifilm, and Amgen. Amgen's campus sits directly across the street from where Genentech is building its facility. This pharmaceutical concentration has fueled rapid local growth, with Holly Springs' population of 48,000 having doubled over the past 16 years.
The expanded Roche investment adds to recent major pharmaceutical commitments in the area, as companies respond to political pressure for increased U.S. manufacturing capacity. Amgen committed $1 billion to its Holly Springs plant in late 2024, while Fujifilm Biotechnologies opened a manufacturing facility in the town last year.
