Rznomics RNA Therapy Shows 23% Complete Response Rate in Liver Cancer Trial
核心洞察
Rznomics (搜索)' RNA-based therapy RZ-001 demonstrated promising interim results in hepatocellular carcinoma (搜索) patients, achieving a 23% complete response rate where tumors completely disappeared.
The clinical trial showed objective response rates of 38.5% by RECIST criteria and 53.8% by mRECIST criteria when RZ-001 was combined with atezolizumab and bevacizumab in treatment-naive patients.
RZ-001 has received FDA orphan drug designation and utilizes RNA trans-splicing technology to cleave telomerase (搜索) mRNA in cancer cells and convert it into therapeutic RNA.
Rznomics (搜索) has reported encouraging interim clinical results for its RNA-based liver cancer (搜索) therapy RZ-001, with approximately 23% of patients experiencing complete tumor disappearance in a Phase I trial targeting hepatocellular carcinoma (搜索). The gene therapy company announced it will present these findings at the American Association for Cancer Research (AACR) conference being held in San Diego from April 17-22.
Novel RNA Trans-Splicing Approach
RZ-001 represents a novel approach to cancer treatment using RNA trans-splicing technology. The therapy works by cleaving telomerase (搜索) messenger ribonucleic acid (mRNA) that appears in cancer cells and converting it into therapeutic RNA to induce cancer cell death. This mechanism targets a fundamental characteristic of cancer cells while potentially sparing healthy tissue.
The drug candidate has received orphan drug designation from the U.S. Food and Drug Administration (搜索) (FDA), providing the company with benefits including priority review, tax credits, waiver of application fees, and up to seven years of market exclusivity upon approval.
Clinical Trial Results
The clinical trial enrolled patients with hepatocellular carcinoma (搜索) who had no prior systemic therapy and for whom transarterial chemoembolization (TACE) was either ineffective or not feasible. RZ-001 was administered in combination with the immune-oncology drugs atezolizumab and bevacizumab, which currently represent the standard first-line treatment for hepatocellular carcinoma.
According to the disclosed abstract, the objective response rate (ORR) reached 38.5% when assessed by RECIST criteria and 53.8% when evaluated using hepatocellular carcinoma (搜索)-specific mRECIST criteria. The complete response rate by mRECIST criteria was 23%, indicating that nearly a quarter of patients experienced complete tumor disappearance.
These response rates appeared higher compared to the combination therapy of atezolizumab and bevacizumab alone, suggesting potential synergistic effects when combined with the RNA therapy.
Company Perspective and Future Plans
Jungwoo Hong, Vice President of Rznomics (搜索), emphasized the preliminary nature of the findings while expressing optimism about the results. "Since additional data has been accumulated after the abstract submission, we will present updated results during the oral presentation," Hong stated. He cautioned that "as these are interim results, we should be cautious about making direct comparisons to other clinical trials or over-interpreting the findings."
The company plans to provide more comprehensive updated clinical results during an oral presentation at the AACR conference on April 19. The additional data accumulated since the abstract submission may provide further insights into the therapy's efficacy and safety profile.
Technology Platform
Rznomics (搜索) has developed proprietary RNA editing technology that can remove RNA causing intractable diseases and express RNA containing therapeutic material. This platform approach suggests potential applications beyond hepatocellular carcinoma (搜索), though the current focus remains on advancing RZ-001 through clinical development for liver cancer (搜索) treatment.
