Novel Trispecific Design Targets Multiple B-Cell Populations

Unlike traditional B-cell therapies, KT501 is engineered to target a broad spectrum of B-cell populations, including plasma cells, by simultaneously binding to CD19 (搜索) and BCMA (搜索). This multi-targeted approach addresses limitations of previous-generation therapies that often spare the cells responsible for autoantibody production.
The trispecific has been developed using Kali's proprietary drug discovery platform, which generates T-cell engagers with precise immune system targeting combined with masking technology designed to avoid off-target side effects. Studies in non-human primates demonstrated strong B-cell depletion capabilities while limiting common side effects of T-cell engagers, including minimizing cytokine release that can cause potentially serious adverse reactions.
Clinical Development Underway
Earlier this month, Kali initiated dosing in a first-in-human Phase 1a trial targeting enrollment of 24 adult subjects with rheumatoid arthritis (搜索).
