Sensory Cloud (搜索) has reported positive results from its Phase II REACH trial of SC0023, a therapeutic aerosol designed to treat refractory chronic cough (搜索) (RCC), conducted at King Saud University Medical City in Saudi Arabia. The study demonstrated a 34% placebo-adjusted cough rate reduction in patients with this challenging condition.
SC0023 contains ions endogenous to human airways and works through daily laryngeal rehydration. The treatment addresses a significant medical need, as chronic cough (搜索) affects an estimated 10% of the adult human population globally, according to REACH trial principal investigator Dr. Kholood Altassan, who described it as "a major unmet healthcare need."
Trial Design and Efficacy Results
The crossover design revealed therapeutic period effects, which limited the primary efficacy analysis to the initial dosing period as a parallel segment design. Post hoc analyses considering investigational product adherence demonstrated the 34% placebo-adjusted cough rate reduction in six participants.
A notable finding was the persistent suppression of cough rates that continued several weeks after dosing, despite SC0023's three-hour endogenous ion clearance time. This extended effect was monitored using Hyfe CoughMonitor watch technology and aligns with treatment decompression as well as down-regulation of mechano-sensitive ion channels (搜索).
Cross-Site Validation and Environmental Factors
Sensory Cloud (搜索) founder and CEO David Edwards explained the rationale for conducting the trial in Riyadh: "By conducting the REACH trial in Riyadh, we wished to explore whether chronic cough (搜索) behaviour and treatment via daily laryngeal rehydration would follow patterns we had previously seen in a recent exploratory trial at Imperial College London, notwithstanding the drier and dustier air in Riyadh."
The results showed that chronic cough (搜索) behavior and treatment efficacy were indistinguishable between the London and Riyadh sites. Edwards noted this "may point to the predominance of time spent within indoor arid-air living environments and the commonality of arid indoor atmospheric conditions."
Next Steps in Development
Based on these positive outcomes, Sensory Cloud (搜索) plans to carry out a follow-up Phase II study at several centers in Belfast, Ireland, and London, UK. This multi-site approach will further validate the therapeutic potential of SC0023 in treating refractory chronic cough (搜索) across different patient populations and geographic locations.
