Safety Profile Supports Dose Escalation
The Phase I/IIa study has successfully enrolled 12 patients across 10 different tumor types, with three patients receiving the current highest dose of 15 MBq/kg. Analysis of data from all treated patients confirms an acceptable and consistent safety profile, leading the DMC to recommend dose escalation to 20 MBq/kg for the next patient cohort.
"It is very gratifying that MTD has not yet been reached and that the DMC recommends a continued increase in dose, as this indicates that Tumorad has an acceptable safety profile so far," said CEO Mats Hansen. "If we maintain the safety profile observed to date, this will be a significant advantage for Tumorad and a clear differentiating factor compared to other RNT drugs, both launched and in development, which often have more complex side effect profiles."
Proof-of-Concept Achieved Through Imaging
The most significant development involves visible tumor uptake of 177Lu-SN201 observed through SPECT imaging in study participants. Particularly notable uptake levels were documented in a patient with adenoid cystic carcinoma (搜索) (ACC) who received one cycle of 177Lu-SN201 at the current highest dose level. The independent monitoring committee considers this observation proof-of-concept for Tumorad, indicating potential therapeutic exposure through delivery of the medically proven isotope 177Lu.
"The tumor uptake demonstrated with 177Lu-SN201 in humans is a significant milestone for the Tumorad program and puts the company in a whole new position," Hansen continued. "The uptake provides important support for the continued development of Tumorad and may open up development paths in indications with the potential for orphan drug status."
Addressing Unmet Medical Need in Rare Cancers
The observed efficacy in adenoid cystic carcinoma (搜索) represents a particularly promising development given the limited treatment landscape for this rare malignancy. ACC is an aggressive cancer (搜索) that most commonly affects the salivary glands and typically manifests between ages 40 and 60. While initial standard treatment involves surgery or radiation, no established standard treatment exists for recurrent or metastatic disease.
Study Design and Next Steps
The Tumorad-01 study represents a first-in-human evaluation designed to assess safety, tolerability, dosimetry, and initial efficacy of 177Lu-SN201 in cancer (搜索) patients. The sequential study design incorporates fixed evaluations by the independent DMC, with the Phase I portion focused on identifying the maximum tolerated dose and potential therapeutic dose for subsequent Phase IIa testing in selected patient populations.
The third patient group, consisting of two men with liver cancer (搜索) and one woman with ACC, has completed treatment with at least one dose cycle of 177Lu-SN201. Based on comprehensive analysis of all available data across the three patient groups, the DMC determined the safety profile remains manageable and consistent, supporting continued dose escalation.
