Sparrow Pharmaceuticals Secures $95 Million Series B to Advance Novel Diabetes Treatment Targeting Elevated Cortisol
核心洞察
Sparrow Pharmaceuticals (搜索) closed a $95 million Series B financing co-led by RA Capital Management (搜索) and Forbion (搜索) to advance clofutriben, a novel HSD-1 inhibitor (搜索) targeting type 2 diabetes (搜索) patients with elevated cortisol (搜索).
Nearly half of patients with difficult-to-control diabetes display increased cardiometabolic risk associated with elevated cortisol (搜索), yet no approved therapies address this underlying etiology for the vast majority of T2D patients.
Clofutriben is a once-daily oral HSD-1 inhibitor (搜索) that modulates intracellular cortisol production in key metabolic tissues and has shown improved glycemic control with favorable safety profile in clinical trials.
Sparrow Pharmaceuticals (搜索) announced the completion of a $95 million Series B financing round to advance its lead candidate clofutriben, a novel HSD-1 inhibitor (搜索) designed to treat type 2 diabetes (搜索) patients with elevated cortisol (搜索). The financing was co-led by RA Capital Management (搜索) and Forbion (搜索), with participation from existing investors including OrbiMed (搜索), RiverVest (搜索), and US Venture Partners (搜索).
Addressing an Unmet Medical Need
The funding targets a significant gap in diabetes care, as nearly half of patients with difficult-to-control diabetes display increased cardiometabolic risk associated with elevated cortisol (搜索). According to Frank Czerwiec, M.D., Ph.D., Chief Medical Officer at Sparrow, "there are no therapies approved for the vast majority of patients with T2D that address this underlying etiology."
Prospective studies have confirmed that excess cortisol drives hyperglycemia (搜索) through multiple mechanisms, including increasing gluconeogenesis, decreasing glucose uptake and disposal, and decreasing insulin synthesis and sensitivity. This multifaceted impact also impairs the effectiveness of standard-of-care treatments, creating a population of patients underserved by current therapies.
Novel Mechanism of Action
Clofutriben represents a once-daily, oral HSD-1 inhibitor (搜索) that modulates intracellular cortisol production in key metabolic tissues. This novel mechanism of action has the potential to complement existing anti-diabetic therapies by directly targeting the causal biology underlying treatment resistance.
In clinical trials conducted to date, clofutriben has demonstrated improved glycemic control, including in individuals with elevated cortisol (搜索), while maintaining a favorable safety and tolerability profile. Notably, the candidate showed no evidence of adrenal insufficiency or requirement for dose titration.
Phase 2b Trial Advancement
The company has advanced clofutriben into the Phase 2b CAPTAIN-T2D trial, a double-blind study evaluating the drug's ability to improve glycemic control and metabolic health in patients with treatment-resistant type 2 diabetes (搜索) and elevated cortisol (搜索). Results from this pivotal trial are expected in 2027.
The CAPTAIN-T2D trial is structured in two parts. Part 1 screens patients with T2D and elevated cortisol (搜索) risk factors for trial eligibility and the presence of elevated cortisol. Eligible participants from Part 1 are then randomized to either clofutriben or placebo in the dose-ranging interventional Part 2. The primary endpoint measures change in hemoglobin A1c (HbA1c) at 24 weeks, with exploratory endpoints including changes in body weight, blood pressure, cholesterol, and bone metabolism biomarkers.
Broader Metabolic Benefits
Beyond glycemic control, HSD-1 (搜索) inhibitors including clofutriben have shown potential in clinical trials for improvement across multiple metabolic parameters. These include weight and body composition, lipids, blood pressure, and markers of bone metabolism, as well as sleep and quality of life improvements, reflecting the broad spectrum of metabolic dysfunction associated with elevated cortisol (搜索).
Strategic Investment and Leadership
In conjunction with the financing, Zach Scheiner from RA Capital Management (搜索) and Nanna Lüneborg from Forbion (搜索) have joined Sparrow's Board of Directors. Robert Jacks, President & Chief Executive Officer, emphasized that "elevated cortisol (搜索) is now recognized as a key driver of disease progression and treatment resistance in millions of patients with type 2 diabetes (搜索) worldwide."
The investment reflects what Jacks described as "a convergence of emerging biological insights, immense clinical opportunity, and payer and clinician receptivity," positioning the funding as potentially enabling "the next big breakthrough in the management of metabolic syndrome (搜索)."
