A Seasoned Clinician-Scientist at the Helm
Dr. Zack is a board-certified rheumatologist with a doctorate in immunology and more than 25 years of experience spanning research, clinical development, and medical affairs. Most recently, she served as chief medical officer at Exagen Inc. Before that, she held clinical leadership positions at Xencor Inc. and within Novartis' Biotherapeutics Development Unit.
Earlier in her career, Dr. Zack served as global development lead for Enbrel at Amgen, where she also held roles across preclinical research, clinical development, and medical affairs. Prior to joining the biopharmaceutical industry, she spent a decade as an associate professor at the University of California, Los Angeles, treating patients with rheumatic diseases while conducting molecular immunology research. She earned her M.D. and Ph.D. from the University of Texas Southwestern Medical School and completed her residency and rheumatology fellowship at the UCLA School of Medicine.
SP001: A Differentiated Approach to Immune-Mediated Disease
Commenting on her appointment, Dr. Zack said SP001 offers a differentiated approach to treating immune-mediated diseases by targeting CD40 ligand, a signaling pathway involved in activating both B cells and T cells. SP001 is a fully humanized, Fc-silent IgG1 monoclonal antibody designed to inhibit CD40L (搜索), an upstream regulator of immune activation involved in T-cell, B-cell, antigen-presenting cell, and platelet biology. Spero is developing the therapy as a targeted, non-B-cell-depleting treatment for immune-mediated diseases where immune cell interactions contribute to chronic inflammation, fibrosis, and disease progression.
The antibody is also designed to address platelet activation concerns associated with earlier generations of anti-CD40L (搜索) therapies while maintaining the extended half-life characteristics of conventional monoclonal antibodies.
