Third Arc Bio Partners with Adagene to Develop Masked CD3 T Cell Engagers for Solid Tumor Treatment
核心洞察
Third Arc Bio (搜索) has entered a licensing agreement with Adagene to develop two masked CD3 (搜索) T cell engagers using Adagene's SAFEbody (搜索) precision masking technology platform.
The partnership aims to address precision challenges in solid tumor immunotherapy by combining Third Arc Bio (搜索)'s ARCStim Platform with Adagene's tumor-specific activation capabilities.
Adagene will receive $5 million upfront and is eligible for up to $840 million in development and commercial milestones, plus royalties on sales.
Third Arc Bio (搜索) and Adagene have announced a strategic licensing agreement that combines two innovative immunotherapy platforms to develop masked CD3 (搜索) T cell engagers targeting solid tumors (搜索). The partnership leverages Adagene's SAFEbody (搜索) precision masking technology with Third Arc Bio's ARCStim Platform to address what executives describe as "one of the biggest challenges in solid tumor immunotherapy: precision."
Financial Terms and Global Rights
Under the agreement terms, Third Arc Bio (搜索) will receive worldwide rights to research, develop, and commercialize two candidate molecules utilizing Adagene's SAFEbody (搜索) technology platform. Adagene will receive an upfront payment of $5 million and is eligible to receive development and commercial-based milestones of up to $840 million if all conditions are achieved, plus royalties on end-user sales.
The deal structure includes a strategic provision allowing Adagene to retain a no-cost option to develop and commercialize these candidate molecules in Greater China, Singapore, and South Korea, providing the company with potential access to key Asian markets.
Technology Integration and Therapeutic Approach
The collaboration centers on Adagene's SAFEbody (搜索) technology, which uses precision masking to shield the binding domain of biologic therapies until activation occurs specifically in the tumor microenvironment. This approach is designed to enable tumor-specific targeting while minimizing on-target off-tumor toxicity in healthy tissues.
"Our SAFEbody (搜索) technology allows activation of an antibody when it reaches the tumor microenvironment," said Peter Luo, Ph.D., Chief Executive Officer of Adagene. "The resulting wider therapeutic index from improved safety allows for higher dosing and potentially better efficacy, as seen from the data we continue to generate with muzastotug."
The technology platform has demonstrated clinical validation through Adagene's lead program, ADG126 (muzastotug), a masked anti-CTLA-4 (搜索) SAFEbody (搜索) currently in Phase 1b/2 and Phase 2 clinical studies in combination with anti-PD-1 (搜索) therapy, particularly focused on microsatellite stable colorectal cancer (搜索).
Expanding T Cell Engager Portfolio
For Third Arc Bio (搜索), the partnership represents an expansion of their existing T cell engager development capabilities. The company's drug development engine focuses on creating multifunctional antibodies that generate immune synapses to precisely activate or inhibit T cells.
"Our agreement with Adagene will allow Third Arc Bio (搜索) to advance highly innovative molecules with a superior therapeutic index and help build on our growing portfolio of novel CD3 (搜索)- and CD28 (搜索)-targeting T cell engagers," said Peter Lebowitz, M.D., Ph.D., Chief Executive Officer of Third Arc Bio. "Adagene's SAFEbody (搜索) technology expands the reach of our ArcStim Platform to additional novel targets."
Third Arc Bio (搜索)'s lead program, ARC101, is a bispecific T cell engager currently being evaluated in a Phase 1 trial for cancer (搜索) patients with solid tumors (搜索) expressing CLDN6 (搜索). The company describes ARC101 as exhibiting high specificity with no off-target binding, avoiding other Claudin family proteins while maintaining potent cytotoxicity and optimal biophysical properties.
Platform Technologies and Future Applications
Adagene's proprietary Dynamic Precision Library platform encompasses NEObody, SAFEbody (搜索), and POWERbody technologies, designed to address safety and tolerability challenges associated with antibody therapeutics. The SAFEbody platform's versatility allows application across various antibody-based therapeutic modalities, including Fc empowered antibodies, antibody-drug conjugates, and bi/multispecific T cell engagers.
Third Arc Bio (搜索) operates two complementary platforms: the solid tumor ARCStim Platform and an immunology & inflammation ARCTag Platform, both designed to offer precision immune regulation for superior efficacy and safety profiles.
The partnership underscores both companies' commitment to addressing the precision challenges inherent in solid tumor immunotherapy, where achieving effective tumor targeting while minimizing systemic toxicity remains a critical therapeutic hurdle.
