TransCode Therapeutics Publishes Breakthrough RIG-I Agonist Study Enabling Tumor-Selective Cancer Immunotherapy
核心洞察
TransCode Therapeutics published preclinical research in Molecular Imaging and Biology demonstrating a novel template-directed RIG-I agonist (搜索) approach that selectively activates innate immune signaling within cancer (搜索) cells.
The innovative strategy uses overexpressed oncogenic microRNAs like miRNA-21 (搜索) as intracellular assembly templates to drive tumor-specific RIG-I (搜索) activation while minimizing systemic toxicity.
The approach addresses longstanding challenges with RIG-I (搜索) agonists including off-target immune activation and inefficient systemic delivery, while enabling non-invasive imaging of drug delivery.
TransCode Therapeutics has published groundbreaking preclinical research demonstrating a novel approach to cancer (搜索) immunotherapy that could overcome major limitations of current RIG-I agonist (搜索) treatments. The study, published February 19, 2026, in Molecular Imaging and Biology, describes a template-directed strategy that enables tumor-selective immune activation while providing real-time imaging capabilities.
Novel Template-Driven Approach Targets Cancer Cells Specifically
The research introduces a template-driven RIG-I agonist (搜索) strategy that selectively activates retinoic acid-inducible gene I (RIG-I (搜索)) signaling inside tumor cells by leveraging overexpressed oncogenic microRNAs, such as miRNA-21 (搜索), as intracellular assembly templates. This innovative approach directly addresses longstanding challenges associated with RIG-I agonists, including off-target immune activation and inefficient systemic delivery.
"Our findings demonstrate a novel approach to precisely engage innate immune pathways directly within tumor cells, while minimizing systemic toxicity," said Zdravka Medarova, Ph.D., CSO of TransCode. "We believe that combining tumor-specific RNA templating with our TTX nanoparticle delivery platform brings RIG-I (搜索)-based immunotherapy closer to clinical relevance."
Addressing Critical Delivery Challenges
The study reports significant improvements over traditional RIG-I (搜索) agonists, demonstrating reduced off-target immune activation and improved targeting capabilities. The template-directed assembly method enables non-invasive imaging of drug delivery, supporting the development of image-guided targeted cancer (搜索) immunotherapy approaches.
The research was conducted in collaboration with Dr. Anna Moore, Professor, Director of the Precision Health Program, and Associate Dean for Research Development at the College of Human Medicine at Michigan State University, who also serves as a scientific co-founder of TransCode.
Clinical Translation Potential
The findings highlight the translational relevance of this approach, particularly given that TransCode's TTX nanoparticle delivery platform is currently being evaluated in clinical trials. The company's lead therapeutic candidate, TTX-MC138, focuses on treating metastatic tumors (搜索) that overexpress microRNA-10b (搜索), a well-documented biomarker of metastasis.
The manuscript, titled "Template-Directed RIG-I Agonist (搜索) Assembly for Image-guided Targeted Cancer (搜索) Immunotherapy," was published in Molecular Imaging and Biology, the peer-reviewed scientific journal and official publication of the World Molecular Imaging Society, which focuses on translational research in molecular imaging and image-guided therapies with clinical and commercial relevance.
