The Series B extension brings together new investors Janus Henderson Investors (搜索), Balyasny Asset Management L.P., and Affinity Asset Advisors alongside existing backers including Alexandria Venture Investments, Eli Lilly and Company, Johnson & Johnson Innovation, Pfizer Ventures (搜索), and others.
Clinical Progress with TRB-061
The company's lead asset, TRB-061, represents a novel approach to treating inflammatory diseases (搜索) through selective activation of regulatory T cells. This purpose-engineered TNFR2 (搜索) agonist is currently being evaluated in a Phase 1a/b clinical trial for moderate-to-severe atopic dermatitis (搜索), with the single ascending dose portion now completed and multiple ascending dose testing underway.
TRB-061 targets tumor necrosis factor receptor 2 (搜索) (TNFR2 (搜索)), a co-stimulatory receptor preferentially expressed on the most suppressive Tregs in skin and gut tissues. The drug is designed to selectively agonize TNFR2, activating tissue-licensed Tregs to address immune-mediated diseases while precisely mimicking the natural TNF ligand.

Addressing Significant Unmet Medical Need
Atopic dermatitis (搜索) affects up to 20% of children and 10% of adults, impacting approximately 204 million people worldwide. The chronic inflammatory skin disease is characterized by rash, itching, and skin barrier dysfunction. In moderate-to-severe cases, patients experience systemic immune activation, extensive body surface area involvement, and substantial quality-of-life impairment.
Current treatment options face significant limitations, with more than half of patients discontinuing existing therapies within two years due to ineffectiveness and long-term side effects. TRexBio (搜索)'s approach aims to reduce inflammation and promote barrier tissue repair by preferentially expanding tissue-licensed Tregs most relevant for immunoregulation, potentially offering superior long-term disease control without broadly suppressing the immune system.
Expanding Pipeline Development
The financing will also support advancement of TRB-071 and TRB-081, two wholly owned preclinical programs with differentiated mechanisms of action designed to address inflammatory and autoimmune conditions (搜索). Both programs emerged from TRexBio (搜索)'s proprietary Deep Biology Platform, which leverages computational and functional analyses of patient samples to understand Treg biology in human tissue. The company expects to initiate Phase 1 clinical trials for each program in 2027.
Competitive Landscape and Market Position
TRexBio (搜索) faces competition from several biotechnology companies developing next-generation eczema treatments, including Corvus Pharmaceuticals, Apogee Therapeutics, and Nektar Therapeutics. Additionally, Sanofi is expected to file for approval of amlitelimab, a follow-up to its blockbuster Dupixent, despite mixed Phase 3 results.
The company's differentiated approach through selective Treg activation represents a potentially transformative therapeutic pillar for long-term inflammatory disease control. Results from the ongoing Phase 1a/b trial of TRB-061 are expected as soon as 2027, which will provide critical data on the safety and efficacy of this novel mechanism.
