TuHURA Biosciences (搜索) announced a $15.6 million equity financing that will fund key milestones across its three-pronged approach to overcoming cancer immunotherapy resistance. The Tampa-based Phase 3 immuno-oncology company is developing novel technologies targeting both primary and acquired resistance mechanisms that limit the effectiveness of current cancer treatments.
The financing provides runway for the company's lead Phase 3 program evaluating IFx-2.0 as adjunctive therapy with Keytruda (pembrolizumab) for first-line treatment of advanced and metastatic Merkel cell carcinoma (搜索). This randomized, placebo-controlled registration trial operates under a Special Protocol Assessment agreement with the FDA and targets completion of enrollment by Q4 2026.
VISTA Emerges as Key Target in AML
A December 5, 2025 mini-symposium featuring key opinion leaders provided scientific rationale for targeting VISTA (搜索) in acute myeloid leukemia (搜索), particularly in combination with menin (搜索) inhibitors for NPM1 (搜索)-mutated relapsed/refractory AML (搜索). The expert panel included Geoffrey Uy from Washington University School of Medicine, Tae Kon Kim from Vanderbilt University Medical Center, and Kevin Yin from Washington University in St. Louis.
"The VISTA (搜索) gene is the only checkpoint upregulated in patients with AML (搜索), notably among high-risk AML," said Dr. James Bianco, President and CEO of TuHURA Biosciences (搜索). "It has been shown that VISTA expression on leukemic blasts is the primary culprit in the low response rate and short duration of response in AML."
The symposium participants noted clear enthusiasm for combining TBS-2025 (搜索), TuHURA's VISTA (搜索)-inhibiting antibody, with menin (搜索) inhibitors across multiple AML (搜索) patient populations, including NPM1 (搜索) relapsed/refractory AML, high-risk AML, and patients unfit for intensive therapies. While menin inhibitors have become standard of care in NPM1-mutated AML, key opinion leaders cited the relatively low complete response rate and short duration of response as significant obstacles requiring new therapeutic approaches.
Preclinical Data Supports VISTA Targeting Strategy
Company studies demonstrated that VISTA (搜索) shows the highest expression in poor-risk AML (搜索) subtypes and is present regardless of common mutations like DNMT3A, NPM1 (搜索), or FLT-3. In murine models of VISTA-expressing AML, TBS-2025 (搜索) provided survival advantages comparable to standard front-line combination chemotherapy while significantly improving survival when used in combination with front-line treatment.
The data showed that inhibiting VISTA (搜索) through either gene silencing or antibody inhibition, combined with menin (搜索) pathway inhibition, significantly improved survival in AML (搜索) mouse models. These findings support the planned randomized Phase 2 trial comparing physician's choice of menin inhibitor versus menin inhibitor plus TBS-2025 (搜索) in NPM1 (搜索)-mutated relapsed/refractory AML, pending FDA clearance.
Delta Opioid Receptor Platform Shows Promise
At the 57th ASH Annual Meeting, TuHURA presented oral and poster presentations positioning the Delta Opioid Receptor (搜索) as a compelling new target for bi-functional, bi-specific immune modulating antibody drug conjugates. The company is leveraging this technology to develop first-in-class ADCs targeting myeloid-derived suppressor cells to inhibit their immune-suppressing effects on the tumor microenvironment.
Dr. Bianco outlined the company's 2026 milestones, including expected completion of Phase 3 enrollment for IFx-2.0 in Merkel cell carcinoma (搜索), preliminary data from the IFx-2.0 basket trial, and data on DOR inhibition effects on MDSCs, TAMs, and T regulatory cells at a Q2 scientific conference. The recent financing builds on TuHURA's $15 million PIPE financing and warrant exercise completed in June 2025. The company completed its merger with Kineta Inc (搜索). on June 30, 2025, acquiring the Phase 2-ready VISTA (搜索)-inhibiting antibody TBS-2025 (搜索).
