The company laid off approximately 90% of its workforce after wrapping up an early-stage clinical trial of VRG50635 in December, according to CEO and founder Alice Zhang. The experimental drug, designed using artificial intelligence technology to treat ALS (搜索), failed to demonstrate therapeutic benefit in patients.
Trial Results Signal Strategic Pivot
VRG50635 was designed to target PIKfyve (搜索), an enzyme involved in cellular waste disposal processes that become impaired in neurodegenerative diseases (搜索) like ALS (搜索). The drug's mechanism was based on research indicating that inhibiting PIKfyve prompts cells to find alternative waste-removal pathways, potentially addressing the toxic protein buildup characteristic of ALS.
However, the trial measured levels of neurofilament light chain (搜索), a protein with known ties to nerve cell damage, and found that these levels increased rather than decreased. This outcome suggested the drug wasn't achieving its intended therapeutic effect.
"A more reasonable expectation is that the technology is going to go through some setbacks, but there's going to be really important learnings that can be fed back into the platform and iteratively used to actually improve on things," Zhang said in an interview with BioPharma Dive.
AI Drug Discovery Challenges
The failure of VRG50635 adds to the growing list of AI-discovered drugs that have struggled to prove their worth in human testing. Other AI-designed compounds, including Recursion Pharmaceuticals' REC-994, have similarly faced challenges in clinical development despite initial promise.
Zhang contextualized these setbacks within the broader pharmaceutical development landscape, noting that failure rates are high even for traditionally discovered drugs. "Never in history has a transformational technology been just built overnight, and it is very rarely the case that the first attempt is the blockbuster," she explained.
New Business Model and Strategic Direction
Under its new identity as Verge Labs (搜索), the company will focus on providing drug developers with data to identify optimal targets for their experimental programs. The business model is "centered around monetizing the datasets and insights," Zhang said, drawing parallels to medtech companies like Tempus AI that have built successful businesses around data services.
The rebranded company will continue its neuroscience focus while partnering with developers to identify new drug targets, find potential patients, and predict new biomarkers. Zhang noted that Verge had considered this strategic pivot for years but that the disappointing trial results finally prompted the company to act.
