Vogenx raises $81.3M in Nasdaq IPO to advance SGLT1 inhibitor mizagliflozin in metabolic disorders
核心洞察
Vogenx (搜索) closed its Nasdaq IPO with $81.3 million raised, surpassing its previously reported $75 million goal, by selling 6,250,000 shares at $13.00 per share.
Proceeds will support development of mizagliflozin, an oral SGLT1 (搜索) inhibitor being evaluated for post-bariatric hypoglycemia (搜索), gastroparesis (搜索), and GIP-dependent Cushing's Syndrome (搜索).
Mizagliflozin is furthest along in post-bariatric hypoglycemia (搜索), where two Phase II trials showed statistically significant improvements in glucose nadir and reductions in hypoglycemic events.
Raleigh, North Carolina-based Vogenx, a clinical-stage biopharmaceutical company, closed its initial public offering Thursday with $81.3 million raised, surpassing its previously reported goal of $75 million. The company sold 6,250,000 shares at $13.00 per share, and its stock trades on the Nasdaq under the symbol VOGX. As of Friday, shares were trading just shy of $15.
The proceeds will support the development of mizagliflozin, an orally administered small molecule that reduces insulin secretion, glucose absorption, and gastric inhibitory peptide secretion. The drug is currently being trialed in post-bariatric hypoglycemia (搜索) (PBH), gastroparesis (搜索), and GIP-dependent Cushing's Syndrome (搜索) (GDCS), with the PBH indication furthest along in Phase IIb.
A gut-targeted SGLT1 mechanism
Mizagliflozin is an oral, minimally absorbed small molecule that selectively inhibits the sodium-glucose transporter 1 (SGLT1 (搜索)) in the intestinal lumen. According to the company, this mechanism reduces and delays glucose absorption and lowers secretion of insulin and glucose-dependent insulinotropic polypeptide (GIP) after meals.
Vogenx (搜索) licensed exclusive global rights to mizagliflozin, excluding Japan, Korea, and Taiwan, from Japan-based Kissei Pharmaceutical (搜索) in December 2021. The agreement requires future development and regulatory milestone payments of approximately $27 million along with a high single-digit royalty on net sales. Kissei retains rights to develop and market the drug in Japan, Korea, and Taiwan.
Vogenx (搜索)'s SGLT1 (搜索)-targeted mechanism differs from SGLT2 inhibitors marketed for type 2 diabetes, which act primarily in the kidney rather than the gut. Competing approaches to post-bariatric hypoglycemia (搜索) remain largely limited to dietary management and off-label use of somatostatin analogs or other glucose-lowering agents, none of which carry FDA approval for the indication.
Clinical progress and pipeline
The company reported completing two Phase II trials of mizagliflozin in PBH patients, in which those dosed with the candidate showed statistically significant improvements in glucose nadir and peak glucose and insulin levels compared with placebo or baseline, along with reductions in hypoglycemic events. Vogenx (搜索) has begun start-up activities for a Phase IIb trial in PBH, called EMERGE, and expects to begin screening patients in 2026.
A Phase II proof-of-concept trial in gastroparesis (搜索) is planned for 2027, subject to additional funding. An investigational new drug application for GDCS, a rare disorder for which the company said no FDA-approved pharmacologic therapies currently exist, is anticipated in 2027, also contingent on further capital.
Vogenx (搜索)'s only other candidate in development is VGX-2857 (搜索), a preclinical SGLT1 (搜索) inhibitor being assessed for weight maintenance after substantial weight loss.
Financing context
Vogenx (搜索) is the smallest IPO among five biotech offerings that entered the Nasdaq in the same week. The others — Braveheart Bio, Latigo Biotherapeutics, Attovia Therapeutics, and BlossomHill Therapeutics — collectively raised more than $1.2 billion, ranging from BlossomHill's $150 million to Braveheart's $382 million. This follows successful fundings from Scribe and Apnimed in July.
The company was founded in 2021 by executives with prior ties to Avolynt and BHV Pharma, including chief executive James Green, chief scientific officer William Wilkison, chief financial officer Steven Delmar, and head of research Bentley Cheatham. Prior to the IPO, Vogenx (搜索) had raised approximately $11.5 million since inception through a Series A convertible preferred stock offering in 2021 and 2022 and a convertible promissory note offering in December 2025.
