Ltd., granting the Chinese biopharmaceutical company exclusive worldwide rights to develop and commercialize HPP737, a novel phosphodiesterase type 4 (PDE4 (搜索)) inhibitor. The amended agreement transforms what was previously a regional partnership into a global collaboration worth up to $135 million in total potential payments.
Strategic Partnership Details
Under the amended license agreement, vTv will receive an immediate upfront payment of $20 million upon execution. The deal structure includes additional milestone payments of up to approximately $50 million for development achievements and up to $65 million in sales milestones, plus tiered royalties based on net sales.

HPP737's Therapeutic Potential
HPP737 represents a potentially significant advancement in PDE4 (搜索) inhibitor therapy for inflammation-mediated diseases, particularly psoriasis (搜索). The compound has demonstrated potent inhibition of interleukin-23 (搜索) (IL-23) and tumor necrosis factor alpha (搜索) (TNF-α) production both in vitro and in vivo, along with therapeutic activity in several animal models of inflammation.
Critically, HPP737 appears to address a major limitation that has historically constrained the therapeutic potential of PDE4 (搜索) inhibitors. In Phase 1 studies, HPP737 was well tolerated with little or no gastrointestinal distress, potentially avoiding the dose-limiting adverse events including nausea and emesis that have limited other PDE4 inhibitors.

Portfolio Context
The HPP737 partnership complements vTv's primary focus on cadisegliatin, a liver-selective glucokinase (搜索) activator currently in Phase 3 trials for type 1 diabetes (搜索). Cadisegliatin has received Breakthrough Therapy designation from the FDA and represents a potential first-in-class oral adjunctive treatment to insulin.
