Zag Bio (搜索) has launched with $80 million in financing to advance what the company describes as the first systemic therapeutics designed to deliver self-antigens directly to the thymus (搜索) for treating autoimmune diseases (搜索). The Cambridge, Massachusetts-based biotechnology company announced its emergence from stealth mode with a Series A financing co-led by founding investor Polaris Partners (搜索) and the T1D Fund (搜索).
The financing round included participation from Mission BioCapital, AbbVie Ventures (搜索), Lightspeed Ventures (搜索), Sanofi Ventures (搜索), KdT Ventures, Regeneron Ventures (搜索), Boxer Capital and Pear VC. Polaris Partners (搜索) founded and incubated Zag Bio (搜索) before co-leading the Series A round.
Novel Thymus-Targeting Mechanism
Zag Bio (搜索) has developed a novel approach to deliver tolerizing antigens to the thymus (搜索) to achieve what the company calls "durable antigen-specific central tolerance" for autoimmune disease treatment. The company designs thymus-targeting bifunctional antibodies that deliver antigens to the thymus, where they engage the body's natural mechanism for T cell tolerance.
This process expands antigen-specific thymic regulatory cells (搜索) (Tregs) while depleting antigen-specific T effector cells (Teff). The thymic Tregs then migrate to diseased tissues to exert protective effects. According to the company, thymic Tregs are epigenetically hardwired to be stable and durable, and are expected to enable an immune reset with long-lasting impact.
"Despite its integral role in central immune tolerance, the thymus (搜索) has been inaccessible to drugs of all types," said Jason Cole, the company's newly appointed Chief Executive Officer. Lead Program Targets Type 1 Diabetes
Zag Bio (搜索) is advancing ZAG-101 (搜索), a bifunctional antibody that delivers pancreatic beta cell antigens to the thymus (搜索), as its lead development candidate for Type 1 diabetes (搜索). The mechanism is designed to expand antigen-specific thymic Tregs to protect beta cells from destruction and preserve their ability to produce insulin in the pancreas.
The company is initially developing ZAG-101 (搜索) for patients who are newly diagnosed with Type 1 diabetes (搜索) or at high risk of developing the disease, with the goal of restoring immune tolerance and preventing autoimmunity. Zag Bio (搜索) has evaluated ZAG-101 in preclinical studies and is initiating IND-enabling studies to advance the program into clinical development in late 2026.
Leadership and Scientific Foundation
The company appointed Jason F. Cole as Chief Executive Officer and Board member. Cole brings over 20 years of biotechnology industry experience, with a track record at public and private companies advancing first-in-class therapies from discovery through approval. His most recent roles included Chief Executive Officer at SalioGen Therapeutics and Chief Strategy & Financial Officer and Chief Business Officer of bluebird bio.
Zag Bio (搜索)'s technology is inspired by the work of co-founder Diane Mathis, Professor of Immunobiology at Harvard Medical School, whose research lab has made pioneering contributions in immunologic tolerance. Other co-founders include John Kulman, the company's Chief Scientific Officer; Jo Viney, a biotech executive with autoimmune disease expertise; and Alan Crane, an entrepreneur partner at Polaris Partners (搜索).

The financing proceeds will support advancing Zag Bio (搜索)'s lead program into the clinic as a tolerogenic therapy to prevent or delay Type 1 diabetes (搜索) onset. Additionally, the funding will enable the company to advance discovery programs for other autoimmune diseases (搜索). Zag Bio's proprietary technology facilitates modular design of thymus (搜索)-targeted self-antigens, enabling streamlined development of additional pipeline programs across multiple autoimmune conditions.
