Inclusion Criteria

1) Must be between the ages of 18-55
2) Schizophrenia patients: must have a diagnosis of schizophrenia or schizoaffective disorder
3) Schizophrenia patients: must be stablized on psychotropic medications
4) Healthy first-degree relative: must have a first-degree relative with schizophrenia or schizoaffective disorder
5) Be mentally competent, as determined by their treating physician.
6) Provide written informed consent.

Exclusion Criteria

1) Non-removable metal objects in head/body
2) pregnancy
3) history of closed or open head injury with loss of consciousness
4) personal history of psychiatric illness (in healthy first-degree relatives and healthy non-relatives)
5) history of neurological illness
6) use of psychotropic medications in healthy controls and relatives
7) substance abuse or dependence within the preceding 6 months (except nicotine)
8) claustrophobia
9) history of schizophrenia in first-degree family members of healthy non-relative controls

Primary Outcome Measures

<p>In general, our main study endpoints will be differences between patients with<br /><br>schizophrenia, healthy first-degree relatives of schizophrenia patients, and<br /><br>demographically-matched healthy controls. Specifically, we are interested<br /><br>group differences for the following measures and their relationships with<br /><br>symptom severity and employment status:<br /><br><br /><br>1. Performance on the search-step task, including efficiency of response<br /><br>inhibition (TSRT) and changes in reaction time as a function of trial history.<br /><br>2. Functional MRI activation measures in several regions of interest<br /><br>3. Differences in structural connectivity between regions of interest using DTI<br /><br>4. Lactate and GABA levels measured using 1H-MRS and their correlations with<br /><br>performance on the search-step task. </p><br>
