Background

A chelating agent (chelating agents) that sequesters a variety of polyvalent cations. It is used in pharmaceutical manufacturing and as a food additive.

Indication

For the reduction of blood levels and depot stores of lead in lead poisoning (acute and chronic) and lead encephalopathy, in both pediatric populations and adults.

Heterologous expression, purification, and biochemical characterization of protease 3075

Protease 3075, a novel enzyme from thermophilic Cohnella sp. A01, was characterized through recombinant production in E. coli BL21. It exhibits optimal activity at 70°C and pH 6, with enhanced stability in the presence of Tween 80 and acetone. Docking and molecular dynamics simulations revealed hydrogen bonds and Van der Waals forces stabilize the enzyme-Tween 80 interaction, increasing structural stability. The enzyme shows potential for industrial applications, particularly in detergents.

C57BL/J6 male mice were used for metabolomics and isotope-labeled MTA analysis under anesthesia. MTA was injected to examine blood–brain barrier crossing. Surgery experiments exposed the left carotid artery, and MTA rescue experiments were conducted. Metabolomics processing involved LC-MS/MS, PCA, PLS-DA, and OPLS-DA. snRNA-seq libraries were generated for single-cell analysis. Joint pathway analysis used O2PLS and MetaboAnalyst 5.0. Western blotting and ELISA assessed protein and cytokine levels. Behavioral tests included open field and Barnes maze tests. A clinical investigation evaluated cognitive decline post-surgery in humans, with serum samples analyzed by LC-MS/MS. Statistical methods included Student’s t-test, ANOVA, and ROC curve analysis.
