The preclinical study, led by CSHL Assistant Professor Corina Amor Vegas in collaboration with Assistant Professor Semir Beyaz, showed significant improvements in gut function, inflammation reduction, and nutrient absorption in both younger and older mice.
Addressing Age-Related Intestinal Decline
Many people experience increased food intolerance with age, which may be attributed to damage in the intestinal epithelium—a thin, single layer of cells essential for digestion and overall gut health. Under normal conditions, this lining renews itself every three to five days. However, aging or exposure to cancer (search) radiation can severely disrupt this renewal process, leading to complications such as leaky gut syndrome (search) and increased inflammation.
The research team applied anti-uPAR (search) CAR T cells directly to the intestines of mice, targeting senescent cells (search) that no longer divide but refuse to die. According to Amor Vegas, the results were striking: "In both cases, we see really significant improvements. They're able to absorb nutrients better. They have much less inflammation. When irritated or injured, their epithelial lining is able to regenerate and heal much faster."
Superior Protection Against Radiation Damage
The study specifically examined the therapy's effectiveness against radiation-induced gut damage, a common complication among cancer (search) patients receiving pelvic or abdominal radiation therapy. Mice exposed to radiation that damaged their intestinal epithelial cells showed remarkable recovery when treated with CAR T cells compared to untreated controls.

Previous studies showed that anti-uPAR (search) CAR T cells could selectively remove senescent cells (search) in mice, leading to major improvements in the animals' metabolism.
The collaboration with Beyaz and graduate student Onur Eskiocak expanded this research to investigate whether removing senescent cells (search) could restore the intestine's healing capacity. The team found compelling evidence that the therapy not only works in animal models but also encourages regeneration in human intestinal and colorectal cells.
