Faeth Therapeutics (搜索), a clinical-stage biotechnology company developing metabolism-based cancer therapies, announced a $25 million strategic financing round to advance its lead PIKTOR (搜索) program following breakthrough clinical results in endometrial cancer (搜索). The funding, led by S2G Ventures (搜索) with participation from existing and new investors, brings the company's total capital raised to $92 million.
Strong Clinical Results Drive Phase 2 Advancement
The financing follows compelling data from Faeth's Phase 1b study of the PIKTOR (搜索) regimen—a combination of serabelisib, sapanisertib, and paclitaxel—which demonstrated an 80% overall response rate in endometrial cancer (搜索) patients. The treatment achieved a median progression-free survival of 11 months, representing a dramatic improvement over the historical 3-4 months seen with chemotherapy alone.
Based on these results, the Gynecologic Oncology Group (GOG) Foundation, one of the premier U.S. clinical trial networks, has initiated a Phase 2 trial (GOG-3111; NCT06463028) that is now enrolling patients. The study includes a predefined substudy testing whether protocolized insulin control through precision nutrition enhances clinical outcomes under trial conditions.
Multi-Node Pathway Inhibition Strategy
Faeth's approach addresses a critical limitation in current cancer therapeutics by targeting the PI3K (搜索)/AKT (搜索)/mTOR (搜索) signaling axis, one of the most frequently altered pathways across solid tumors including endometrial, breast, lung, and ovarian cancers. Unlike conventional single-node inhibitors that often fail due to feedback reactivation and pathway mutations, Faeth employs selective multi-node inhibition at PI3K⍺, mTORC1 (搜索), and mTORC2 (搜索) while controlling the nutrient supply that tumors depend on.

In preclinical models, this multi-node approach achieved more complete pathway shutdown than single-agent inhibition. Multi-node inhibition of the PI3K (搜索)/AKT (搜索)/mTOR (搜索) pathway has already shown clinical benefit in Phase 3 studies, confirming the value of pathway-level control in both mutant and wild-type tumors.
MetabOS Platform Expansion
The funding will expand throughput for Faeth's MetabOS™ platform, an AI-driven system that integrates genomic, gene expression, and tumor microenvironment data to identify metabolic dependencies and exploit them clinically. The platform recognizes that tumors have significantly elevated metabolic demands and require specific nutrients to survive, enabling Faeth to simulate and validate more regimens designed to address metabolic escape.

Pipeline Expansion Beyond Oncology
The financing supports advancement of the Phase 2 endometrial cancer (搜索) program through a full data readout in Q3 2026, while also enabling expansion into additional indications. The funding will support a Phase 1 study in locally advanced rectal cancer (搜索) for Faeth's non-essential amino acid restricted program and initiation of the company's first non-oncology program in Hereditary Tyrosinemia Type 1 (搜索) (HT1), a rare pediatric metabolic disorder affecting 1 in 100,000 births.
For HT1, where toxic metabolites accumulate due to enzyme deficiency and current treatments still leave unmet medical needs despite preventing liver failure, Faeth is applying MetabOS to develop metabolic therapeutic approaches. IND-enabling studies are targeting Q4 2026 clinical entry, representing the first demonstration of MetabOS applied outside oncology.
Investment Perspective

Clinical data from sapanisertib in combination with paclitaxel, one component of the PIKTOR (搜索) regimen, have been selected for a late-breaking oral presentation at the ESMO 2025 Congress.
