Novel Sequential Treatment Strategy
The BLAZE trial will investigate Faron's lead asset bexmarilimab in combination with Gilead Sciences' anti-PD-1 (search) checkpoint inhibitor zimberelimab. The study targets patients with metastatic NSCLC and melanoma who have developed resistance to prior anti-PD-1-based immunotherapy and progressed within three months of their last anti-PD-1 treatment.
The trial employs an innovative approach by first priming patients with bexmarilimab monotherapy, followed by the combination of bexmarilimab and zimberelimab. This sequential strategy aims to resensitize tumors to anti-PD-1 (search) treatment after disease progression.

Addressing Critical Unmet Medical Need

While checkpoint inhibitors have revolutionized cancer therapy, the majority of patients with advanced melanoma and lung cancer eventually experience disease progression. The scientific rationale for the BLAZE trial centers on evidence that tumor-associated macrophages (TAMs) create an immunosuppressive tumor microenvironment that drives resistance to checkpoint inhibitors. Bexmarilimab is designed to overcome resistance to existing treatments by targeting myeloid cell function and activating the immune system. The drug binds to Clever-1 (search), an immunosuppressive receptor found on macrophages that contributes to tumor growth and metastases by helping cancer evade immune surveillance.
By targeting the Clever-1 (search) receptor, bexmarilimab reprograms macrophages, upregulates interferon production, and primes the immune system to attack tumors while sensitizing cancer cells to standard of care treatments. Zimberelimab, the combination partner, is an anti-PD-1 (search) monoclonal antibody that binds PD-1 with the goal of restoring antitumor T cell activity.
