Compass Therapeutics Advances Multiple Bispecific Antibody Programs with Key Biliary Tract Cancer Data Expected Q1 2026
核心洞察
Compass Therapeutics reports confirmed third response with CTX-8371 in Hodgkin lymphoma (搜索) patient, expanding therapeutic potential beyond solid tumors to hematologic malignancies.
The company's tovecimig Phase 2/3 COMPANION-002 study in biliary tract cancer (搜索) remains on track for key survival data in late Q1 2026, following successful primary endpoint achievement.
CTX-8371 cohort expansions are now enrolling patients with triple-negative breast cancer (搜索) and non-small cell lung cancer (搜索) based on durable responses observed in Phase 1 studies.
Compass Therapeutics has provided a comprehensive corporate update highlighting significant progress across its pipeline of bispecific antibody therapeutics, with multiple data readouts expected throughout 2026. The clinical-stage oncology company is advancing first-in-class treatments targeting critical pathways in cancer biology, with particular momentum in biliary tract cancer (搜索) and checkpoint inhibitor-resistant malignancies.
Tovecimig Shows Promise in Biliary Tract Cancer
The company's lead candidate tovecimig, a DLL4 (搜索) x VEGF-A (搜索) bispecific antibody, continues to advance through the Phase 2/3 COMPANION-002 study in patients with advanced biliary tract cancer (搜索). Key secondary endpoint analyses for progression-free survival and overall survival remain on track for late Q1 2026, following the successful achievement of the primary endpoint announced in April 2025.
The COMPANION-002 trial demonstrated that tovecimig in combination with paclitaxel achieved a 17.1% overall response rate, including one complete response, compared to 5.3% for paclitaxel alone in second-line biliary tract cancer (搜索) patients. This represents a significant advancement for a cancer type with less than 20% five-year survival rates.
"Key survival analyses for tovecimig in late Q1, if positive, could transform the treatment of patients with biliary tract cancer (搜索)," said Thomas Schuetz, MD, PhD, Chief Executive Officer and Vice Chairman of the Board of Directors.
Tovecimig has also demonstrated activity in colorectal cancer (搜索), with Phase 2 monotherapy data showing a 5% overall response rate in heavily pre-treated patients who had all received prior bevacizumab treatment. This activity differentiates tovecimig from other anti-VEGF agents, which have shown 1.0-1.5% response rates in this patient population. The data will be presented at the 2026 ASCO GI Cancers Symposium.
CTX-8371 Expands Into Hematologic Malignancies
The company's PD-1 (搜索) x PD-L1 (搜索) bispecific antibody CTX-8371 has achieved a significant milestone with confirmation of a third response in a patient with Hodgkin lymphoma (搜索). This response expands the therapeutic potential beyond solid tumors to hematologic malignancies, prompting investigation of accelerated pathways for both adult and pediatric Hodgkin lymphoma patients in the post-checkpoint inhibitor setting.
In solid tumors, CTX-8371 continues to show durable responses, with a previously disclosed deep partial response in a triple-negative breast cancer (搜索) patient remaining durable through week 32. Based on these encouraging results, cohort expansions are now open for enrollment in patients with triple-negative breast cancer and non-small cell lung cancer (搜索), with 28 patients planned for each indication.
Pipeline Expansion and Leadership Strengthening
Compass has expanded its pipeline with CTX-10726 (搜索), a PD-1 (搜索) x VEGF-A (搜索) bispecific antibody expected to enter Phase 1 studies in Q1 2026. Preclinical data demonstrated superior tumor control compared to ivonescimab in head-to-head studies using human NSCLC xenograft mouse models, with initial clinical data expected in the second half of 2026.
The company has strengthened its leadership team with the appointment of Arjun Prasad as Chief Commercial Officer and Cynthia Sirard, MD, as Chief Medical Officer. Prasad brings experience in successful oncology product launches, including targeted therapeutics for biliary tract cancer (搜索) patients, while Dr. Sirard contributes deep oncology clinical development expertise.
Financial Position and Future Outlook
With an estimated $209 million in cash and marketable securities as of December 31, 2025, Compass maintains a cash runway extending into 2028. This financial position supports the company's ambitious development timeline, which includes multiple data readouts and potential regulatory filings throughout 2026.
"We are advancing multiple first-in-class, bispecific antibodies and expect to achieve major data-driven milestones this year," Schuetz noted. The company anticipates presenting dose-escalation data for CTX-8371 following the biliary tract cancer (搜索) results, with potential for filing its first Biologics License Application for tovecimig later in the year.
