Daily Probiotic Shows Modest Benefit for Depression and Anxiety in Seniors: Pilot Trial

The pilot study, published in the Journal of the American Geriatrics Society, enrolled 58 adults aged 60 and older with moderate depression in India, all of whom continued their regular antidepressant regimens throughout the trial.
Participants were randomly assigned in a 1:1 ratio to receive either a daily probiotic supplement or a placebo for 12 weeks, followed by an additional 12-week monitoring period to assess longer-term outcomes. Both groups demonstrated substantial improvement over the course of the study. However, those in the probiotic group experienced somewhat greater reductions in symptoms of depression (搜索) and anxiety (搜索) compared with those receiving placebo.
Biological Markers and the Gut-Brain Axis
The study went beyond psychological rating scales to examine biological underpinnings of the observed effects. Researchers measured serum brain-derived neurotrophic factor (BDNF), a protein critical for the growth, maintenance, and survival of nerve cells that is frequently studied in mental health research. They also conducted fecal microbiota profiling to analyze the composition of gut microbes in participants.
The findings lent support to the gut-brain connection hypothesis—the idea that the trillions of microbes residing in the human digestive tract may influence mood, behavior, and mental health through various biological pathways. Probiotics, as live microorganisms that can help support a healthy bacterial balance in the gut, are being investigated as potential complements to traditional psychiatric treatments.
Modest Gains, Not a Panacea
While the results are encouraging, the researchers did not find clear evidence that probiotics produced additional improvements in overall quality of life compared with placebo. The benefits, though measurable on depression (搜索) and anxiety (搜索) scales, were modest in magnitude.
Because the study was a relatively small pilot, the findings should be viewed as preliminary. Larger studies will be needed to determine how much benefit probiotics may provide, which patients are most likely to respond, and whether the effects remain consistent across broader populations.

Even so, the results support the idea that probiotics could serve as a safe and biologically plausible addition to standard depression treatment, particularly for older adults who may be seeking complementary options alongside conventional pharmacotherapy.
